淫羊藿和女贞子对激素干预哮喘大鼠模型HPA轴及糖皮质激素受体的影响

摘　　要：目的:研究哮喘大鼠模型地塞米松干预撤停不同阶段(撤停前、撤停中、撤停后)合用中药淫羊藿、女贞子对下丘脑-垂体-肾上腺(HPA)轴及糖皮质激素受体(GR)的影响。方法:卵蛋白致敏、激发建立大鼠哮喘模型,腹腔注射地塞米松模拟激素干预过程,分别在激素干预不同阶段(撤停前、撤停中、撤停后)合用中药淫羊藿、女贞子,比较药物影响激素干预各阶段哮喘模型支气管肺泡灌洗液(BALF)中GR蛋白表达,血清皮质醇(COR)、血浆促肾上腺皮质激素(ACTH),下丘脑促肾上腺皮质激素释放激素(CRH)含量。结果:激素干预组在地塞米松撤停各阶段均出现COR含量显著降低,ACTH含量显著增高。BALF中糖皮质激素受体(GR)蛋白结合力下调(P<0.05,P<0.01),激素撤停前阶段CRH含量上升;而淫羊藿女贞子合激素干预各组中,以淫羊藿女贞子合激素干预Ⅱ组对COR及GR上调作用最佳,并可抑制ACTH的增高。结论:激素撤停中阶段合用中药淫羊藿、女贞子对糖皮质激素作用途径(HPA轴及GR的含量与功能)保护作用最佳。Objective： To observe the effects of the compatibility of Epimedium and Ligustrum lucidum on HPA axis and GR of asthmatic in rats with glucocorticoid （GC） intervention at different treatment stage （before withdrawal, during the withdrawal and after withdrawal）. Methods： Dexamethasone was injected into asthmatic model in rats with the oral gavage of Epimedium and Ligustrum lucidum at different treatment stages of GC withdraw. The following testing indexes, including the content of serum COR, plasma ACTH, hypothalamic CRH and BALF GR, were observed. Results： At the different periods of Dexamethasone withdrawal, serum COR content and BALF C,-C-GR binding capacity in GC group were significantly lower （P〈0.05, P〈0.01）; II groups of the combination of Herba Epimedii, Ligustrum lucidum and GC could increase the content of COR in serum and the GC-GR binding capacity in BALF significantly, compared with the GC group. Conclusion： During the period of GC withdrawal, the combination of Herba Epimedii and Lignstrurn lucidum has a better protective effect on glucocorticoid pathways （including HPA axis and GR）.
