摘  要:目的:研究黑蒜提取液联合放疗对荷H22小鼠移植瘤生长的影响,并初步探讨其分子机制。方法:建立荷H22小鼠皮下移植瘤模型,对4组移植瘤模型分别给予0.9%氯化钠溶液、γ射线照射、黑蒜提取液及黑蒜提取液联合γ射线照射的处理,2周后摘取眼球取血后脱臼处死小鼠。剥离瘤体,计算肿瘤生长抑制率;流式细胞术Annexin V-FITC/PI双染法检测肿瘤组织细胞凋亡情况;免疫组织化学法检测肿瘤组织中Bax及Bcl-2蛋白的表达情况;紫外分光光度计法检测各组小鼠血清中SOD及CAT的活力变化。结果:黑蒜提取液联合放疗可显著抑制荷H22小鼠移植瘤的生长,肿瘤生长抑制率高达66.93%;并能够明显诱导H22细胞凋亡,凋亡率可达(53.49±1.98)%,且上调移植瘤组织中Bax及下调Bcl-2蛋白的表达;黑蒜提取液联合放疗可显著提高小鼠血清中SOD和CAT的活力(P<0.05),增强小鼠抗氧化能力。结论:黑蒜提取液联合放疗可明显抑制荷H22小鼠移植瘤的生长,其作用机制可能与其下调抗凋亡Bcl-2蛋白及上调促凋亡Bax蛋白的表达和提高机体抗氧化能力有关。Objective: To explore the effects on tumor growth with H22 by the combination of black garlic extract and radiotherapy and preliminary discuss the possible molecular mechanism. Methods: The tumor-bearing H22 mouse modle was established, 40 mice were randomized into 4 groups: control group (peritoneal injection 0.9%NS, 0.2mL), irradiation group ( peritoneal injection 0.9%NS, 0.2mL+yray, 2Gy/day), black garlic extract group(peritoneal injection, lg/mL black garlic extract 0.2mL), Black garlic extract + irradiation group (peritoneal injection, lg/mL black garlic extract 0.2mL +Tray, 2Gy/day). After two weeks, mice were sacrificed after eye blood had been gathered, then the tumors were separated and weighed to calculate the tumor-inhibiting rate. Flow cytometry Annexin V-FITC/PI was used to detect tumor cell apoptosis. Immunohistochemistry methods were used in detecting tumor Bax and Bcl-2 protein expression. Ultraviolet spectrophotometer method was used to detect the vigor change of SOD and CAT in mice serum. Results: Black garlic extract combined with radiotherapy could significantly increase the tumor-inhibiting rate, the tumor-inhibiting rate reached as high as 66.93%. The combination of black garlic extract and radiotherapy significantly induced H22 cell apoptosis with the apoptosis rate (53.49+1.98)%, upregulating the expression of Bax and downregulating the expression of Bcl-2 (P〈0.05). Black garlic extract combined radiotherapy could enhance the antioxidant ability by improving the mice serum SOD and CAT vigor. Conclusion: Black garlic extract combined radiotherapy can obviously increase the tumor-inhibiting rate of H22 mice. The effect is probably mediated by upregulating the expression of Bax and dowrtregulating the expression of Bcl-2. It also proves to be associated with oxidation resistance.
