摘 要:目的:研究91个炎症细胞因子基因在内毒素休克大鼠脑组织动态差异表达情况。方法:动物选用体质量180-200g的6-8周龄SD大鼠48只,随机分为空白对照1、2、3、6h组与模型1、2、3、6h组,每组6只。建立内毒素休克大鼠模型,首次采用PCR芯片技术,检测并分析内毒素休克大鼠脑组织1、2、3、6h不同时间段的基因表达情况,统计方法采用△△Ct。结果:①芯片实验结果,模型组与空白对照组比较得到差异表达基因67个,占总基因的73.6%。差异表达基因在1、2、3、6h不同时间有动态变化。按生物学功能分类:信号转导34个,炎症应答36个,G蛋白偶联受体蛋白信号转导途径20个,细胞凋亡11个,细胞增殖15个,趋化性33个,调控转录2个,钙离子稳态7个。②ELISA法测定IL-1α结果,模型组与空白对照组比较1h组明显下降(P<0.01),2h组、3h组无显著性差异,6h组明显升高(P<0.01),并且在6h时模型组IL-1α含量为4个时间段的最高值,与芯片变化规律基本相同。结论:通过对大鼠脑组织细胞因子基因表达谱的构建,筛查出与内毒素休克脑损伤密切相关的细胞因子有趋化因子配体7(Ccl7)、趋化因子配体11(Ccl11)、趋化因子配体9(Ccl9),并且发现主要的信号转导途径有G蛋白偶联受体蛋白信号转导途径。Objective: To study 91 cytokine genes dynamic different expression in cerebral tissue of rats with endotoxic shock. Methods: 48 SD rats were randomly divided into blank control group, model group. Endotoxic shock model was induced and PCR chip technology was firstly used to detect genes expression.of cerebral tissue of rats with endotoxic shock at 1, 2, 3 and 6 hours by means of AACt. Results: QCompared to blank control group, gene expression of 67 genes in rats endotoxic model were changed, accounting for 73.6%, among them the changing expression of genes was regulated dynamically at 1, 2, 3 and 6 hours. According to functional classification of biology: 34 genes related with signal transduction, 36 genes inflammatory response, 20 genes G protein coupled receptor signaling pathways, 11 genes apoptosis, 15 genes cell proliferation, 33 genes chemotaxis, 2 genes regulation and transcription, 7 genes Ca:+ steady. (2)Compared to blank control group, IL-lu level of model group was lower at 1 hour (P〈0.01), no obvious changes at 2 and 3 hours, higher at 6 hour (P〈0.01). IL-la level in gene expression and protein expression showed the same tendency. Conclusion: Constructing the gene expression profile of cytokines in cerebral tissue of rats with endotoxic shock can find out Cc17, Ccll 1, Cel9 related with endotoxic shock and the main signal pathway, G-protein coupled receptor kinase protein signal transduction pathway.
