摘 要:以丝素和壳聚糖为原料,采用冷冻干燥法制备丝素/壳聚糖支架,通过预矿化使羟基磷灰石(HA)沉积,形成多孔羟基磷灰石-丝素蛋白/壳聚糖复合支架。对复合支架进行红外吸收光谱(FT-IR)、X射线衍射(XRD)、扫描电子显微镜(SEM)、热重分析(TGA)、力学性能和空隙率表征;采用WST-1方法检测MG-63骨肉瘤细胞在复合支架上的增殖情况,评价其细胞相容性。结果显示,沉积的HA结构属于弱结晶态范畴,主要位于孔壁上,且随着预矿化循环次数的增加,沉积量增多,使预矿化复合支架的弹性模量得到极显著的提高,其孔隙率保持在90%左右的较高水平;WST-1检测表明,含有HA的预矿化复合支架对细胞增殖有明显的促进作用,具有良好的细胞相容性。Silk fibroin/chitosan scaffolds were prepared through lyophilization, then hydroxyapatite was deposited by premineralization to obtain porous hydroxyapatite(HA)-silk fibroin/chitosan composite scaffolds.Composite scaffolds were characterized by Fourier transform infrared analysis(FT-IR), X-ray diffraction(XRD), scanning electronic microscopy(SEM), thermogravimetry analysis(TGA), compressive modulus and porosity,as well as the proliferation of MG-63 cells on composite scaffolds was detected by WST-1 method. The results reveal that the structure of deposited HA is a weak crystallinity, which deposits on the wall of pore. With the increase of premineralization cycles,the amount of HA and compressive modulus of premineralized composite scaffolds are significantly increased, the porosity keeps about 90%. WST-1 assay shows that premineralized composite scaffolds deposited with HA dramatically enhance the proliferation of cells, and possess a great cytocompatibility.
