摘 要:目的探讨丹参酮ⅡA对大鼠心肺复苏后脑NMDAR1蛋白表达的影响及对脑组织的保护作用。方法将78只SD雄性大鼠随机(随机数字法)分为3组:A组(假手术组,n=6),B组(对照组,n=36),C组(丹参酮ⅡA干预组,n=36),通过制备窒息致大鼠心脏骤停动物模型,C组复苏即刻予以丹参酮ⅡA15mg/kg静脉推注,B组予以等量生理盐水静脉推注,在自主循环恢复后(ROSC)1、6、12、24、48、72h断头取脑,应用免疫组织化学染色法检测脑组织NMDAR1和Caspases-3的表达,干湿质量法测脑组织含水量。实验结果组间比较采用单因素方差分析。结果①B组在ROSC后1h脑NMDAR1蛋白表达明显增高,6h达到高峰,12h逐渐降低,24h仍高于正常,在48h表达低于正常,72h更低,与A组比较差异具有统计学意义(P〈0.05);C组1、6、12hNMDAR1蛋白的表达与同时间点B组比明显降低(P〈0.01),但24、48、72hNMDAR1蛋白的表达差异无统计学意义(P〉0.05)。②B组Caspases-3蛋白表达在ROSC后呈上升趋势,48h达高峰,后逐渐降低,但72h仍高于正常,与A组比较差异具有统计学意义(P〈0.01);C组在1、6、12、24h明显低于B组(P〈0.01),48h、72h表达仍低于B组(P〈0.05)。③B组大鼠脑组织含水量在ROSC后1h就开始升高,24h达高峰,48h逐渐降低,但72h仍高于正常,与A组比较差异具有统计学意义(P〈0.01或P〈0.05);与B组比较,C组含水量明显降低(P〈0.01)。结论丹参酮ⅡA对大鼠心肺复苏后早期NMDAR1的表达有下调作用,能明显减少复苏后脑Caspases-3的表达及脑水肿。Objective To investigate the impact of tanshinone Ⅱ A on the level of brain NMDAR1 protein in rats after cardiopulmonary resuscitation and its effects of brain function protection. Methods Seventy-eight SD male rats were randomly (random number) divided into three groups: group A (sham group, n = 6), group B ( control group, n = 36) and group C ( Tanshinone Ⅱ A intervention group, n = 36). All animals were induced to be models of cardiac arrest by choking. The rats of group C received intravenous injection of Tanshinone Ⅱ A in dose of 15 mg/kg immediately at initiation of resuscitation, while rats of group B were intravenous injected same amount of normal saline instead. Brains tissues of all rats were taken at 1, 6, 12, 24, 48 and 72 h after the restoration of spontaneous circulation (ROSC). Immunohistochemical staining method was applied for measuring the levels of brain tissue NMDAR1 and Caspases-3, while water content of the brain was detected by wet and dry weight ratio. The experimental data were analyzed by using one-way ANOVA. Results ①The level of brain NMDAR1 protein in group B increased at 1 h and reached its peak at 6 h after ROSC, then its level gradually declined and dropped below normal at 48 h, 72 b, and there were significant difference in variation of NMDAR1 protein levels in comparison with the group A ( P 〈 0. 05 ) ; the NMDAR1 protein levels at 1, 6, 12 h in group C were significantly lower than those in group B at the same intervals ( P 〈 0.01 ), but no significa/at differences were seen at 24, 48, 72 h ( P 〉 0.05 ). ②The level of brain Caspases-3 in group B increased after ROSC, and reached its peak at 48 h after ROSC, then declined and maintained above normal at 72 h, and this variation was significantly different from that of the group A (P 〈0.01 ) ; while the levels of caspase-3 at 1, 6, 12, 24 h in group C were significantly lower than those in group B (P 〈0. 01), but thses differences at 48, 72 h were still significant ( P
