摘  要:目的研究慢性激动G蛋白偶联受体30(GPR30)对大鼠心力衰竭的保护作用。方法40只SD大鼠随机均分为假手术组(A组)、卵巢切除组(B组)、异丙肾上腺素85mg·kg-1·d-1模型组(C组)、GPR30激动剂G-1 120μg·kg-1·d-1给药组(D组)、雌激素40μg·kg-1·d-1给药组(E组)。记录左心室收缩压(LVSP)、左心室舒张末压(LVEDP)、左心室压变化最大速率(±dp/dtmax),检测各组心肌梗死面积及心肌细胞凋亡。结果与A组、B组相比,C组大鼠LVDP和±dp/dtmax降低,LVEDP升高,心肌梗死面积增大,凋亡心肌细胞数量增多(P<0.01或P<0.05),而D组、E组能改善上述指标的变化过程(P<0.05)。结论慢性激动GPR30可对抗心肌细胞凋亡,从而对大鼠心力衰竭产生保护作用。Objective To investigate the protective effect of chronic activation of G protein- coupled receptor 30 (GPR30) on rats with heart failure. Methods Forty SD rats were equally randomized into five groups of A(sham operated), B(ovariotomy model), C(injected with isoproterenol 85 mg- kg-1· d-1 ),D(injected with GPR30 agonist G-1 120 ·μg. kg-1 · d-1) and E(injected with 17β-estradiol 40 vg · kg-1 · Kg-1 ). Left ventricular systolic pressure(LVSP), left ventricular end diastolic pressure(LVEDP) and maximal rate of left ventricular pressure(±dp/dtmax) were recorded. The area of myocardial infarction was calculated and apoptosis of myocardial cells was determined by TUNEL method. Results Compared with groups of A and B, LVSP and dp/dtmax were decreased, while the LVEDP, area of myocardial infarction and apoptosis of myocardial cells were significantly increased in group C(P〈0. 01 or P〈0. 05),which were all obviously improved in groups of D and E (P〈0. 05). Conclusion Chronic activation of GPR30 has cardioprotective effect on rats with heart failure probably via inhibiting apoptosis of myocardial cells.
