摘  要:目的探讨脑源性神经生长因子(BDNF)对皮层神经元谷氨酸释放的作用及可能机制。方法体外分离培养SD大鼠皮层神经元,用Src特异性抑制剂PP2 10μmol/L预处理30min后,再加入BDNF 100ng/ml刺激。采用比色法检测皮层神经元谷氨酸释放,免疫印记检测Src、磷脂酶C-γ1(Plc-γ1)、细胞外调节蛋白激酶(ERK1/2)和Akt的表达变化。结果 BDNF能促进谷氨酸释放,增加Src、Plc-γ、Akt和ERK1/2的磷酸化水平。而PP2降低BDNF刺激皮层神经元的谷氨酸释放,抑制Plc-γ1的磷酸化水平;但对ERK1/2和Akt的磷酸化水平无明显影响。结论 BDNF可能通过Src/Plc-γ1信号通路刺激皮层神经元的谷氨酸释放。Objective To investigate the effect and underlying mechanism of brain-derived neurotrophic faetor(BDNF) on the release of glntamic acid from cortical neurons. Methods The cortical neurons of SD rats were isolated and cultured in vitro. After pretreated with Src inhibitor PP2 10 μmol/L for 30 min, the cortical neurons were stimulated with BDNF 100 ng/ml. The release of glutamic acid from cortical neurons was deteimined by colorimetry, and the expressions of Src, Plc-γ1, ERK1/2 and Akt were detected by Western blot. Results BDNF significantly promoted the release of glutamic acid and simultaneously increased the phosphorylation levels of Src, Plc-γ1, Akt and ERK1/2. However, PP2 decreased BDNF-stimulated release of glutamic acid, inhibited the phosphorylation level of Plc-yl, but had no effect on the phosphorylation levels of ERK1/2 and Akt. Conclusion BDNF can stimulate the release of glutarnic acid from cortical neurons probably via Src/Plc-γ1 signaling pathway.
