补肺颗粒对慢性阻塞性肺疾病稳定期模型大鼠血清炎性因子水平的影响

摘　　要：目的:观察补肺颗粒对慢性阻塞性肺疾病(COPD)稳定期模型大鼠肺组织病理学改变及血清炎性因子水平的影响,初步探讨其疗效作用机制。方法:将60只雄性Wistar大鼠随机分为两组,空白对照组24只,COPD稳定期模型组36只。采用熏香烟结合气道内滴注内毒素法制造COPD稳定期大鼠模型,造模结束后,两组各随机抽取12只取材;而后将模型组剩余24只随机等分为两组,即空白治疗组和补肺颗粒干预组。空白对照组及空白治疗组给予生理盐水灌服,干预组给予补肺颗粒治疗,均持续30天。观察指标:①苏木色精-伊红法染色(HE染色)观察肺组织病理学改变;②酶联免疫吸附法(ELISA)检测大鼠血清白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)及高敏C反应蛋白(hs-CRP)含量。结果:造模后,模型组大鼠肺组织可见慢性支气管炎及阻塞性肺气肿的病理特点,IL-6、IL-8、TNF-α及hs-CRP含量较空白对照组显著升高(P<0.05)。补肺颗粒治疗后,肺组织病变较前减轻,治疗后3组间比较,干预组IL-6、TNF-α、hs-CRP含量明显降低(P<0.05);自身治疗前后比较,干预组IL-8、TNF-α、hs-CRP含量显著降低(P<0.05)。结论:COPD稳定期体内存在以血清炎性因子普遍升高为特点的微炎症状态,补肺颗粒可降低其水平,改善肺组织病变,减轻炎症反应,是有效的治疗药物。Objective：To observe the effect of Bufei Granules on the pathological changes（ PC ）of the lung tissue and the levels of the serum inflammatory factors （SIF）in rats with chronic obstructive puhnonary disease （COPD）stable phase （SP） , and to explain the mechanism of its therapeutic effect preliminarily. Methods ：60 male Wistar rats were randomly divided into the control group （CG, n = 24）and the COPD SP model group（ MG, n = 36）. We made COPD SP rat model by giving cigarette smoke to them and injecting the lipopolysaccharide （LPS）into their airways. After that, we selected 12 rats from the two groups respectively to draw materials. Then the 24 rats remained in the MG were divided into the control treatment group（CTG） and the Bufei Granules intervention group（BGIG） randomly. The rats in the CG and the CTG were given by saline, and the rats in the BGIG were treated by Bufei Granules solution. Treatment lasted for 30 days. Outcome measures：（1） the PC of the lung tissue （ HE staining） ;（2） detection of the levels of serum interleukin-6 （ IL-6 ）, interleukin-8 （ IL-8 ）, tumor necrosis factor-α（TNF-α） and high-sensitivity C-reactive protein（hs-CRP） by the method of enzyme-linked immunosorbent assay （ELISA）. Results：We could find the pathological features of chronic bronchitis and emphysema in pathological slices of the lung tissue in the MG, and the levels of IL-6, IL-8,TNF-α and hs-CRP in the MG were higher than those in the CG（ P 〈 0.05 ）. After treatment,the PC in the BGIG mitigated, and the levels of IL-6, TNF-α and hs-CRP lowered significantly（ P 〈 0.05 ） in the comparison of the three groups. The levels of IL -8, TNF-α and hs-CRP in the BGIG after treatment were conspicuously lower than before （ P 〈 0.05 ）. Conclusion ： The micro-inflammatory state characterized by the elevation of the SIF levels is certified to exist in patients with COPD SP. Bufei Granules is an effective drug, which can lower the levels of the SIF
