Wortmannin对糖尿病肾病大鼠肾组织TNF-α与VEGF表达的影响

摘　　要：目的研究糖尿病肾病(DN)大鼠模型经静脉注射PI3K抑制剂Wortmannin后肾组织肿瘤坏死因子-α(TNF-α)与血管内皮生长因子(VEGF)表达的变化。方法成年健康清洁级SD雄性大鼠30只,随机分为假手术组(10)、模型组组(10)和Wortmannin处理组。参照Anderson′s方法制作糖尿病肾病大鼠模型。采用Western blot方法检测三组大鼠肾组织TNF-α与VEGF蛋白的表达;RT-PCR方法检测三组大鼠肾组织TNF-αmRNA与VEGFmRNA的表达。结果 Western blot结果显示,模型组与Wortmannin处理组肾组织TNF-α与VEGF蛋白的表达量均显著高于假手术组(P<0.05),而与模型组相比,Wortmannin处理组肾组织TNF-α与VEGF蛋白的表达量明显降低(P<0.05);RT-PCR结果显示,与假手术组比较,模型组与Wortmannin处理组肾组织TNF-αmRNA与VEGFmRNA的表达量均显著升高(P<0.05),而Wortmannin处理组肾组织TNF-αmRNA与VEGFmRNA的表达则明显低于模型组(P<0.05)。结论 PI3K抑制剂Wortmannin能够下调DN大鼠肾组织TNF-α与VEGF的表达。Objective To study the expressions of tumor necrosis factor nuclear factor-α（TNF-α） and vascular endo- thelial growth factor （VEGF） in diabetic nephopathy rat kidney after Wortmanninon was injected. Methods 30 healthy male rats were selected and randomly divided into the sham group（10）, the model group（10）and Wortmannin treatment group. The diabetic nephropathy model was induced with Anderson＇s way. The expressions of TNF-a and VEGF were observed by western blot method in all groups;The expressions of TNF-α mRNA and VEGFmRNA were observed by RT-PCR method in all groups. Results The expressions of TNF-α and VEGF increased significantly in model group and Wortmannin treatment group than in sham group by means of western blot（P〈0.05） ,but decreased in Wortman- nin treatment group than in model group （P〈01 05） ;Compared with the sham group,the expressions of TNF-α mRNA and VEGFmRNA were increased significantly in model group and Wortmannin treatment group （P〈0. 05）, but de- creased in Wortmannin treatment group than in model group （P〈0.05）. Conclusion Wortmannin could down-regulate the expressions of TNF-α and VEGF.
