摘  要:目的通过应用基因敲除(APOE-/-)小鼠的动脉粥样硬化斑块的模型进一步探讨免疫反应在动脉动脉粥样硬化的作用。方法建立小鼠动脉粥样硬化硬化模型。实验小鼠选取Apoe基因敲除(Apoe-/-)小鼠,及与基因敲除小鼠同源的C57BL/6小鼠,并分别随机分为高脂饮食组(Ah、Ch),正常饮食组(An、Cn);Ah组及Ch组小鼠给予高脂饮食喂养12周后处死动物,切取主动脉弓并观察粥样斑块形成情况。主动脉标本做HE染色,光镜下观察病理改变;使用免疫组织化学染色方法检测粥样斑块中CD8+T淋巴细胞的含量;应用Motic Images Advanced3.2图像分析仪系统测量主动脉内膜CD8+T免疫沉积物平均灰度值。结果动脉粥样硬化斑块大体形态:Ah组小鼠有动脉粥样硬化斑块形成,斑块主要集中在动脉分叉处。An组小鼠无动脉粥样硬化斑块形成。Ch组小鼠未见动脉斑块形成,但腹腔及各组织脏器表面脂肪含量明显增多。Cn组小鼠无动脉斑块形成。HE染色标本中,Ah组斑块程度、泡沫细胞数量较Cn组明显增多,Ch组较正常对照组斑块程度、泡沫细胞数量增多,但程度较轻,An组与正常对照组相比,内膜光滑,无明显差别。Ah组、Ch组及An组小鼠表达CD8+T细胞较Cn组明显减少(P<0.01);CD8+T表达程度表现为:Ah<Ch<An<Cn。结论在小鼠动脉粥样硬化模型中随着斑块数量、泡沫细胞数目增多,CD8+T淋巴细胞的表达呈减少趋势,CD8+T淋巴细胞与动脉粥样硬化进展的免疫机制相关。Objective through the application of gene knockout (APOE - / -) mice atheromatous plaque model fur- ther discussed immune response in the artery atherosclerosis(AS) rote. Methods to establish AS hardening model in mice. I.aboratory mice select Apoe knockout (Apoe-/-) mice,and knockout mice homologous C54BL / 6 mice,two groups of mice were randomly divided into high fat diet group (Ah,Ch) ,normal diet group (An,Ch) ; Ah group and Ch mice fed ahigh-fat diet to 12 weeks. 12 weeks after death animals,chipped aortic arch and observe atheromatous plaque formation conditions. Aortic specimen for HE staining,then observed the pathological change,the use of immunohisto- chemical staining method detection in this atheromatous plaque CD8 -6 T lymphocytes content, application of Motic Im- ages Advanced3.2 image analyzer system measuring aortic intima CD8 -6 T immune deposits average gray value. Re- sults The results of AS plaque general form:Ah mice have atheromatous plaque formation,plaque mainly concentrated in the artery crotch, plaque quantity medium. An mice without AS plaque formation. Ch mice did not see artery plaque formation,but the abdominal cavity and the organization organ surface fat content increased obviously. Cn mice without artery plaque formation. HE dyed samples, Ah group plaque degree, foam cell number is a significant increase in Cn group,Ch group than normal control group plaque degree, foam cell number increase, but to a lesser degree, An group and normal control group,compared to intima smooth,no obvious difference. Ah group,Ch group and An mice express CD8+T cells is Cn group significantly reduced,(P 〈0.01) ; CD8+ T express degree performance:Ah 〈 Ch 〈 An Cn. Conclusion in mice atherosclerosis model with plaque quantity, foam cell populations,CD8+T lymphocytes expression decreased,CD8+T lymphocytes and progression of atherosclerosis immune mechanism related.
