摘  要:目的本研究旨在建立一种快速、灵敏、特异的抗生素残留检测方法。方法通过选取两种分别能与氯霉素、卡那霉素特异性结合的适配分子,把含有不同浓度氯霉素、卡那霉素的模拟样品结合于硝酸纤维膜上,通过条件的优化,建立基于适配分子的Dot-ELISA快速检测氯霉素、卡那霉素残留的方法。结果该方法可以快速检测牛奶等食品中的氯霉素、卡那霉素残留,检测灵敏度达10-7g/L。结论与现有的氯霉素、卡那霉素残留检测方法相比,基于适配分子的Dot-ELISA方法可快速检测氯霉素、卡那霉素残留,且具有灵敏性高、特异性强等特点,可根据这两种抗生素的检测方法与条件为基础,延伸到其他抗生素残留的检测,可作为抗生素残留筛查的较好选择,有广阔的应用前景。Objective To establish a fast,sensitive and specific method to detect antibiotic residues.Methods The aptamers which could specifically combined with chloromycetion and kanamycin were selected.In the aptamer assay,the samples containing different concentrations of chloromycetion and kanamycin were spotted on the nitrocellulose (NC) membrane with optimization.The NC membrane was incubated with biotin-labeled chloromycetion specific aptamer at 55℃ for 1 hour and then washed.The membrane was incubated with streptavidin-labeled HRP and colored by DAB.Results The assay could rapidly detect chloromycetion and kanamycin residues in milk and other foods.A concentration as low as 10 7 g/L could be detected with linearity.Conclusions Compared with the existing assay for detecting chlommycetion and kanamycin residues,.the method presented here is sensitive and specific.It will be a good choice in antibiotics screening with broad prospects in application.
