摘  要:目的:探讨雷公藤内酯醇(triptolide)诱导骨髓增生异常综合征(MDS)细胞系SKM-1细胞凋亡及机制。方法:Triptolide与SKM-1细胞共育,观察细胞形态,流式细胞术检测细胞凋亡,RT—PCR检测Bcl-2、Bax与caspase-3基因表达。结果:40—160ng/mLtriptolide明显促进SKM.1细胞凋亡,随triptolide浓度增加及时间延长,凋亡发生率增加(P〈0.01),Bcl-2表达下降(P〈0.01),Bax、caspase-3表达上升(P〈0.01)。结论:40~160n/mL-triotolide对SKM-1细胞有促凋亡作用.可能通过下调Bcl-2、上调Bax、caspase-3基因表达起作用.Objective To investigate the mechanism of triptolide inducing the apoptosis of myelodysplastic syndrome (MDS) cell line SKM-1. Methods SKM-1 cells were incubated with triptolide, and then the cellular morphology was observed, flow cytometry analysis was used to detect the apoptosis, RT-PCR was used to detect the expressions of Bcl-2, Bax and caspase-3. Results 40-160 ng/mL of triptolide could obviously promote the apoptosis of SKM-1 ceils. With the increase of the acting time and concentration of triptolide, the apoptosis rate was increased (P 〈 0.01), the expression of Bcl-2 was decreased (P 〈 0.01), and the expressions of Bax and caspase-3 were increased (P 〈 0.01 ). Conclusion 40-160 ng/mL of triptolide may promote the apoptosis of SKM-1 cells through down-regulating the expression of Bcl-2 and up-regulating the expressions of Bax and caspase-3.
