摘  要:目的:探讨犬冻干骨与骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)的相容性,研究其作为骨组织工程支架材料的可行性。方法:杂种犬下颌骨行骨块制备、深冻、脱脂、冷冻干燥、灭菌制成冻干骨。分离培养同种异体犬BMSCs,并与冻干骨复合培养。HE染色观察冻干骨的组织结构。MTT比色法检测冻干骨对BMSCs的活性影响。扫描电镜观察细胞在冻干骨表面的生长情况。结果:冻干骨对BMSCs的活性无影响。扫描电镜观察可见细胞紧密黏附冻干骨生长,细胞形态呈多角形。结论:所制备犬冻干骨能与BMSCs具有良好的细胞相容性。Objective To evaluate the cellular compatibility of canine freeze-dried bone (FDB) to bone marrow mesenehymal stem cell (BMSCs), and to verify the feasibility of canine FDB as scaffold material in bone tissue engineering. Methods Canine FDB was made through block preparation, deep frozen, degreasing, freeze drying and sterilization. Allogenetic BMSCs were cultured on FDB in vitro. The tissue structure of FDB was observed by histochemistry staining. The influence of FDB on the proliferation of BMCSs was observed by MTT method. The adhesion and growth of BMSCs were observed by scanning electron raicroscope (SEM). Results There was no influence of canine FDB on the proliferation of BMSCs. Under the SEM, the BMSCs were tightly adhered on the surface of the canine FDB, grew well and showed polygon shape. Conclusion The prepared canine FDB had good cytocompatibility to BMSCs.
