摘 要:为研究烟酰胺(nicotinamide,NA)对小鼠脂肪细胞能量代谢的影响,本试验添加0,100,200,300,400,500μmol/L NA干预24 h后,检测小鼠脂肪细胞一氧化氮(NO)生成量和三磷酸腺苷酶(ATPase)活性,利用实时荧光定量PCR检测细胞过氧化物酶体增殖物激活受体基因(PPARγ)的表达。结果表明:随着NA浓度的增加,NO生成量减少,脂肪细胞能量代谢提高,PPARγ基因mRNA表达受到明显抑制,其中以200μmol/L NA的效果最为显著(P<0.01),但500μmol/L NA反而抑制了细胞的能量代谢。表明在一定浓度范围内,烟酰胺具有促进脂肪细胞能量代谢的作用,这可能对于肥胖、脂肪肝等多种脂肪能量代谢相关疾病有一定的治疗作用。To investigate the effect of nicotinamide (NA) on energy metabolism of mice adipocytes in vitro,adipocytes were cultured with 0,100,200,300,400,500 μmol/L NA for 24 h,then nitrogen oxide (NO) concentration in culture medium and intracellular adenosine triphosphatase (ATPase) activity were assessed,and mRNA expression of the peroxisome proliferator-activated receptor gamma gene (PPARγ) was analyzed using real-time PCR.Results showed that with the increase of the concentration of NA (0 ~200 μmol/L),NO concentration reduced,while energy metabolism of mice adipocytes was improved and PPARγ mRNA expression was markedly inhibited,compared with the control group (P 〈 0.01).However,energy metabolism was inhibited with higher concentration of NA (500 μmol/L).The results above indicated that NA could promote energy metabolism of mice adipocytes in vitro within a certain concentration,which may have a therapeutic effect for obesity,fatty liver and other fat energy metabolism related diseases.
