摘  要:目的观察四磨汤对寒冷束缚应激小鼠促胰液素(Sec)和P物质(SP)mRNA表达的影响,揭示四磨汤调节胃肠功能紊乱的作用机制。方法动物随机分组,采用寒冷束缚等多种刺激方法,制备胃肠功能紊乱小鼠模型,分别给予蒸馏水(模型组)、得舒特(对照组)、四磨汤(四磨汤组)治疗,用RT-PCR法检测结肠组织Sec、SP mRNA表达。结果模型组小鼠结肠组织中Sec mRNA的表达下降、SP mRNA表达上升,与正常组比较差异有统计学意义(P<0.01);四磨汤组Sec mRNA的表达增加、SP mRNA表达下降,与模型组比较差异有统计学意义(P<0.05)。结论四磨汤治疗功能性胃肠病的作用机制可能与调节Sec及SP mRNA的变化有关。Objective: To study the effects of Simo decoction on expression of Sec and SP mRNA in cold-con- straint stressed mice,and to explore the mechanism of Simo decoction on FGIDs. Methods: Gastrointestinal dys- function 'model was established by exerting the factors of cold-constraint stimulation. And then randomly treated with distilled water(model group), Dicetel (control guoup) and Simo decoction (SD group). The expressions of SEC and SP in intestine were measured by RT-PCR. Result: The expressions of SEC mRNA of model group was de- creased,and SP mRNA of model group was increased, compared with the normal group (P〈 0.01). The SD could increase the expression of SEC mRNA in intestine. The average optical density of immune positive result has sig- nificant differences compared with the model group(P〈 0.05). The Simo decoction could reduce the expression of SP mRNA in intestine,which also has significant differences compared with the model group (P〈 0.05).Conclu- sion: Simo decoction appears potential to regulate the expression of SEC and SP mRNA in cold-constraint stressed mice,which is one of possible mechanism to therapy FGIDs.
