芍药苷对血吸虫肝纤维化小鼠肿瘤坏死因子α基因及蛋白水平的影响

摘　　要：目的从血清、基因、蛋白表达水平探讨小鼠感染血吸虫后肝纤维化不同时期给予芍药苷治疗对肝组织肿瘤坏死因子α(TNF-α)的影响。方法除正常组(Ⅳ组)外,将BALB/C小鼠随机分为杀虫前(Ⅰ组)、杀虫同时(Ⅱ组)及杀虫后给药组(Ⅲ组),Ⅰ组于感染12 d开始给予芍药苷和溶剂媒,Ⅱ组于感染42 d给药,Ⅲ组于感染72 d给药,均用药30 d,所有感染小鼠于42 d以吡喹酮杀虫,用药结束后60 d处死小鼠,取血和肝脏。采用放免法、RT-PCR法、Western-blotting法和免疫组化法分别观测血清及肝组织中TNF-α转录和表达情况。结果芍药苷可显著降低血清及mRNA水平的TNF-α表达;而在Western-blotting法和免疫组化法中,仅在Ⅰ组和Ⅲ组,芍药苷明显降低TNF-α的含量。结论TNF-α与肝纤维化呈正相关。芍药苷可抑制虫卵肉芽肿,减少胶原生成,TNF-α水平也相应降低。Objective To investigate the effect of paeoniflorin（PAE） on the expression level of tumor necrosis factor-α （TNF-α） from the serum,gene and protein expression levels in treating different periods of mice hepatic fibrosis after infection by Schistosoma japon- icum. Methods In addition to the normal group（ IV ）,the BALB/C mice were randomLy divided into the pre-disinsection treatment group （I）,disinsection simultaneous treatment group （Ⅱ） and post- disinseetion treatment group（ Ⅲ ）. PAE and solvent media were given from 12 d after infection in the group Ⅰ,from 42 d ＇after infection in the group Ⅱ and from 72 d after infection in the group Ⅲ, the medication lasted for 30 d. All infected mice were given oral praziquantel（PZQ） on 42 d after infection for disinsection and killed on 60 d after medication for collecting blood and liver. The transcription and expression of TNF- OL in blood and liver tissues were detected by the radioimmunoassay, RT- PCR, Western blotting and immunohistochemical methods. Results PAE apparently de- creased the TNF- α expression level of serum and mRNA;in Western blotting and immunohistochemical tests,PAE obviously decreased the TNF- α expression level in the group I and Ⅲ. Conclusion TNF- α is positively correlated with hepatic fibrosis. PAE can sup- press egg granuloma,reduce the generation of collagen and correspondingly decrease the TNF- α level.
