摘  要:目的探讨不同浓度丹酚酸B(salvianolic acid B,SA-B)对小鼠体外血小板P-选择素的作用。方法 50只雄性昆明小鼠随机分为5组:对照组、模型组、低浓度SA-B组、中浓度SA-B组,高浓度SA-B组,眼球摘除采血。对照组加入生理盐水,其余各组加入血小板活化剂胶原(终浓度为100μg/L),再将不同浓度的SA-B加入经胶原预处理的小鼠全血中(SA-B的终浓度分别为400mg/L,800mg/L及1200mg/L),采用流式细胞技术测血小板膜P-选择素阳性率。结果模型组P-选择素高于对照组(P<0.01);低浓度SA-B组、中浓度SA-B组、高浓度SA-B组P-选择素均低于模型组(P<0.05);不同浓度SA-B组P-选择素随SA-B浓度的增加而减少(P<0.05)。结论 SA-B能抑制胶原引起的血小板活化,且随着浓度的增加,该抑制作用更加明显。Objective To study the effect of different concentrations of salvianolic acid B (SA - B)on platelet P - selectin in mice. Methods Fifty male KM mice were randomly assigned to 5 groups : Control group, model group,low concentration of SA - B group, middle concentration of SA -B and high concentration of SA -B group, 10 in each group. Blood sample was collected by removing the eyeballs. The platelet was activated by collagen firstly,and then different concentration of SA- B were added into the mouse blood. P - selectin was detected by the method of flow cytometry. Results P - selectin in model group was higher than that in control group(P^0.01). Com- pared with model group,P - selectin significantly decreased in low concentration of SA - B group, middle concentration of SA - B and high concentration of SA - B group (P%0.05). P - selectin in different concentrations of SA - B groups decreased with the increasing concentration of SA - B(P〈0.05). Conclusion SA - B can inhibit the activation of platelet induced by collagen, moreover,the inhibi- tory effect enhances with the increasing concentration of SA - B.
