摘 要:目的:探讨核转录因子红细胞系-2相关因子2(Nrf2)在小鼠缺氧性肺动脉高压病程中的作用及其表达趋势。方法:健康成年C57小鼠40只,雌雄不限,随机分为正常对照组(n=10)、缺氧1周组(n=10)、缺氧2周组(n=10)、缺氧4周组(n=10)。采用缺氧性肺动脉高压模型。Western blot检测Nrf2在小鼠缺氧性肺动脉高压病程中的表达;检测肺动脉形态学和右心室形态改变;同时记录右心室血流动力学变化。结果:随缺氧时间的增加,右心室收缩压逐渐增高,缺氧4周后右心室平均收缩压较之于对照组显著增高(P<0.05);肺小动脉血管壁逐渐增厚,管腔狭窄,至缺氧4周管腔进一步狭窄,较之于对照组增加显著(P<0.05);右心室和左心室加室间隔的比值[RV/(LV+S)]随缺氧时间的延长,其增幅亦更加显著(P<0.05)。小鼠肺动脉平滑肌细胞Nrf2的表达随着慢性缺氧的时间增加亦逐步增高(P<0.05)。结论:Nrf2在缺氧性肺动脉高压病程进展中起重要作用,但其具体机制仍待进一步的研究阐明。Objective: To investigate the dynamic expression of Nrf2 during the course of pulmonary ar- terial hypertension (PAH) in mouse. Methods: Forty C57 mice were divided equally and random- ly into four groups: the normal control group (group C, n=10), hypoxia 1-week group (group HlW, n=10), hypoxia 2-week group (group H2W, n=10), and hypoxia 4-week group (group H4W, n= 10). PAH model was established, and then the expression of Nrf2 during the course of PAH in mouse was measured by Western blot. Morphology of pulmonary artery and right ventri cle was observed, and systolic blood pressure of right ventricle was detected at the same time. Results: Systolic blood pressure of right ventricle in group H4W were significantly increased with the increasing of anoxic time (P〈0.05). The thickness in pulmonary artery increased gradually (P〈0.05) and the ratio of right ventricle to left ventricle plus septum increased dramatically af- ter 4 weeks' hypoxia (P〈0.05). The expression of Nrf2 in mouse pulmonary arterial smooth muscle cell progressively increased with the course of PAH (P〈0.05). Conclusion: Up-regula tion of Nrf2 expression plays pivotal role during the course of pulmonary arterial hypertension,but the specific mechanism is still waiting for study.
