摘 要:目的:观察黄连解毒汤(Huanglian Jiedu Decoction,HJD)对东莨菪碱致记忆获得性障碍模型小鼠学习记忆能力及中枢胆碱能系统的影响。方法:将60只小鼠按体重、性别随机分为6组,分别为空白对照组、东莨菪碱模型组、安理申阳性组(2.05mg/kg)、黄连解毒汤低(3g/kg)、中(6 g/kg)、高(12 g/kg)剂量组。连续灌服受试药物7天后造模,正常对照组和东莨菪碱模型组每日灌服等剂量生理盐水,除正常对照组腹腔注射等剂量生理盐水外,其余5组小鼠腹腔注射东莨菪碱(3mg/kg)建立记忆获得性障碍模型。30 min后进行Morris水迷宫实验,测试结束后处死动物,摘全脑在低温环境下分离大脑皮层、海马组织,于匀浆管中充分研碎,制成10%脑组织匀浆,比色法检测AchE活性、ChAT活性。结果:与模型组比较,黄连解毒汤各剂量组均可提高模型小鼠的学习记忆能力,缩短逃避潜伏期时间(P<0.05),延长有效区停留时间及游泳路程(P<0.05),并能降低模型小鼠脑内AchE活性,提高ChAT活性(P<0.05);与安理申阳性组比较,黄连解毒汤各剂量组除ChAT活性,其他指标均无差异。结论:黄连解毒汤可改善东莨菪碱致记忆获得性障碍模型小鼠的学习记忆能力,其机制可能与增强ChAT活性、抑制AchE活性有关。To observe the effects of Huanglianjiedu Decoction (HID) on Learning-Memory Ability and Cholinerglc System of Memory Disorder Model Induced by Scopolamine in Mice. Methods: According to the weight and gender, 60 mice were randomly divided into six groups, including blank control group, model group, positive control group(2.05mg/kg), HID low-dose group, middle-dose group, high dose group(3,6,12g/kg). After 7 d administration, acquired memory disorder model was made with scopolamine subcutaneous injection in the dose of 3mg/kg. After 30 rain mice were subjected to Morris water maze test. the activity of acetyleholinesterase (AchE) and acetylcho line transferase (CHAT) in cerebral cortex was measured by a biochemical method. Results: Compared with model group, a significant de crease of escape latency and increase of swimming time, swimming distance in effective area were observed in HID groups( P 〈 0.05). HID decrease AchE activity and increase ChAT activity in the cerebral cortex of model mice ( P 〈 0.05). Compared with positive control group, there is no significant difference among all indexes in HID groups except ChAT activity. Conclusions: HID effectively improved learning- memory ability in model mice induced by scopolamine, and the mechanism might be related with the decrease of the activity of AchE and the increase of the activity of CHAT.
