摘 要:目的:观察慢性温和不可预知应激(CUMS)模型大鼠胃肠功能的异常变化及逍遥散的影响。方法:运用慢性温和不可预知应激结合孤养方式制备CUMS大鼠模型,造模2周后灌胃给予逍遥散(20g/kg、30g/kg)和阿米替林(10mg/kg)进行治疗,连续给药4周。于造模前、造模2、6周测定大鼠糖水消耗率,于造模6周测定大鼠小肠推进率和胃排空率,ELISA法测定大鼠血清胃泌素、胃动素含量。结果:与正常组比较,模型组大鼠的糖水消耗率明显降低,小肠推进率及胃排空率均显著性下降,血清胃泌素、胃动素含量下降。与模型组比较,逍遥散30g/kg能对抗模型大鼠糖水消耗率的降低,对模型大鼠胃肠运动的异常具有对抗作用,并表现出提高模型大鼠血清胃泌素、胃动素水平的趋势。结论:CUMS结合孤养方式制备的抑郁症模型大鼠胃肠功能呈现一定紊乱表现,逍遥散具有对抗作用,作用机制可能与影响胃肠激素的分泌有关。To observe the change of gastrointestinal motility in CUMS model rats, and the effects of xiaoyao powder on it. Methods: The depression model was replicated by chronic unpredictable mild stress and single cage feeding in SD rat. Model rats were administrated with Xiaoyao Powder(20g/kg,30g/kg) and amitfiptyline( 10mg/kg) for 4 weeks after being accepted stress for 2 weeks. The consumption of sweet solution of rats were measured at different time, and small intestine driving action and gastric emptying rates of model rats were meas ured. The contents of gastrin and motilin in serum in rats were detected by ELISA assay. Result: Compared with the normal group, the con sumption of sweet solution was decreased in model group, as well as the small intestine pushing rates and gastric emptying rates were de creased. The gastrin and motilin contents in serum were also decreased in model group. Compared with the model group ,Xiaoyao powder could increase the consumption of sweet solution, reverse the abnormal changes of gastrointestinal motility, and exhibit the increasing trend on the level of gastrin and motilin in serum. Conclusion:The gastrointestinal motility of CUMS model rat showed the disorder, Xiaoyao powder exhibi ted the amendment influence on this abnormal changes, which the mechanism be related to regulating the secretion of gastrointestinal hormones.
