左旋延胡索乙素对化疗痛大鼠痛阈的影响及其在脊髓水平的机理研究

摘　　要：目的:通过观察左旋延胡索乙素(L-THP)对长春新碱诱导的神经病理性疼痛大鼠痛阈值及其脊髓腰段TNF-α、IL-1β表达的影响,探讨L-THP对化疗痛的作用及其在脊髓水平的作用机制。方法:根据痛阈值大小筛选出合格的大鼠30只,并随机分为5组:对照组、模型组、L-THP 3mg/kg、6mg/kg、12mg/kg组。采用隔日腹腔注射长春新碱(125μg/kg)的方法建立化疗痛动物模型,采用不同剂量的L-THP治疗化疗痛大鼠,并测定大鼠机械痛阈值和热痛阈值,用ELISA方法检测脊髓腰段TNF-α、IL-1β含量,比较各组的变化。结果:与对照组比较,模型组机械痛阈值和热痛阈值明显降低(P<0.05或P<0.01);与模型组比较,L-THP12mg/kg组和L-THP 6mg/kg组机械痛阈值或热痛阈值显著增高(P<0.05或P<0.01)。另外,模型组TNF-α、IL-1β水平与对照组组比较显著升高(P<0.01),L-THP 12mg/kg组和L-THP 6mg/kg组TNF-α、IL-1β表达较VCR组明显降低(P<0.05或P<0.01)。结论:左旋延胡索乙素能提高化疗痛大鼠的痛阈值,该作用可能与其抑制脊髓腰段TNF-α、IL-1β等炎性细胞因子的表达有关。Observing effects of Levo-tetrabydropalmatine （L-THP） on pain threshold and the expressions of TNF-α and IL-1βin the spi nal cord in rats with vineristine-induced neuropathic pain, to investigte the role of L-THP and spinal mechanisms. Methods： 30 SD rats were randomly divided into 5 groups： control group , model group , L-THP group（3mg/kg,6mg/kg,12mg/kg）. Vincristine （125p.g/kg, i. p. ） was administered to rats on alternate days to create chemotherapy-induced neuropathie pain model in rat, then treated with different dose of L- THP. Pain threshold and expressions of TNF-αand IL-1 β were determined respectively. Results： Compared with control group, the pain threshold decreased in model group （ P 〈 0. 05 or P 〈 0.01 ） ; Compared with model group, the pain threshold increased in L-THP group （6mg/kg or 12mg/kg）（P 〈0.05 or P 〈 0.01 ）. Moreover, compared with control group, the expression of TNF-α and IL-lβincreased in model group （P 〈 0.01 ） ; Compared with model group, the expression of TNF-α and IL-1 βdecreased in L-THP group（6mg/kg, 12mg/kg） （P 〈 0.05 or P 〈 0.01 ）. Conclusion： L-THP can relieve the chemotherapy-induced neuropathic pain, probably by attenuating the expression of TNF-α and IL-1 β in spinal cord.
