摘 要:目的:观察荆防散乙酸乙酯提取物的抗过敏作用,为其临床应用及抗过敏作用机制研究提供实验依据。方法:采用DNCB诱发小鼠迟发型超敏反应实验,组胺、5-HT致小鼠毛细血管通透性增加实验,右旋糖酐致小鼠瘙痒实验及小鼠同种、异种被动皮肤过敏反应实验,观察荆防散乙酸乙酯提取物10.01g/kg、6.67g/kg、3.34g/kg(分别为成人每日用药量的30、20、10倍)的抗过敏作用。结果:荆防散乙酸乙酯提取物6.67g/kg、10.01g/kg连续灌胃给药9天,显著降低DNCB诱发的小鼠迟发型超敏反应所致的耳肿胀,并降低胸腺、脾脏指数;能明显抑制组胺所致的小鼠皮肤毛细血管通透性增高;6.67g/kg连续灌胃给药9天,显著抑制5-HT所致的小鼠皮肤毛细血管通透性增高。荆防散乙酸乙酯提取物10.01g/kg连续灌胃给药7天,明显缩短右旋糖酐所致的小鼠瘙痒反应持续时间;6.67g/kg、10.01g/kg连续灌胃给药6天,显著降低小鼠同种、异种被动皮肤过敏反应中小鼠腹部皮肤毛细血管通透性的增高。结论:荆防散乙酸乙酯提取物具有良好的抗过敏作用,进一步验证了该部位为荆防散抗过敏作用的有效部位。To observe the anti-allergic effects of the ethyl acetate extract of Jingfang Powder, so as to provide experimental evidence for its clinical use and further studies of the anti-allergy mechanism. Methods: Six kinds of allergic reaction model in mice were used to investi gate the anti-allergic effects of the ethyl acetate extract, which including the model of DNCB-indueed delayed-type hypersensitivity in mice, and histamine or 5-HT induced increased capillary permeability of abdominal skin in mice, and dextran induced pruritus reaction in mice, and the homologous or heterologous passive cutaneous anaphylaxis in mice. The dosage of the ethyl acetate extract of Jingfang Powder respec tively was 10.01g/kg, 6.67g/kg and 3.34g/kg. Results: The ethyl acetate extract of Jiugfang Powder were intragastricly administrated for nine days at the dose of 6.67g/kg and 10.01g/kg, could significantly reduce the ear swelling in the DNCB-induced delayed-type hypersensi tivity in mice, while reduced the index of thymus and spleen ( P 〈 0.05, P 〈 0.01 ), and also significantly inhibited the increasing of abdomi nal skin capillary permeability induced by histamine in mice ( P 〈 0.05). At the dose of 6.67g/kg, it could significantly inhibit the skin cap illary permeability induced by 5-HT in mice after continuous administration for nine days ( P 〈 0.05 ). The ethyl acetate extract of Jing-fang powder were intragastricly administrated for seven days at the dose of 10.01g/kg, was able to shorten the duration of pruritus response in duced by dextran ( P 〈 0.05). The ethyl acetate extract were intragastricly administrated for six days, under the dose of 6.67g/kg, 10.01 g/ kg significantly reduced the increasing of abdominal skin capillary permeability in homologous or heterologous passive cutaneous anaphylaxis in mice ( P 〈0.05 ,P 〈0.01 ).
