摘  要:目的利用二氮嗪(DZ)预处理骨骼肌成肌细胞,探讨DZ预处理对H2O2损伤L6骨骼肌成肌细胞的作用。方法培养L6骨骼肌成肌细胞株(L6SkM),分成正常对照组、H2O2损伤组和DZ预处理组,用HE染色、MTT、LDH检测、免疫组化、流式等方法检测DZ预处理对氧化应激损伤L6骨骼肌成肌细胞的作用。结果 DZ预处理能改善氧化应激损伤L6骨骼肌成肌细胞的形态结构,减少细胞凋亡,明显提高细胞的生存率。结论 DZ预处理对氧化应激损伤L6骨骼肌成肌细胞具有明显的保护作用。Objective To explore the protective effects of diazoxide (DZ) on I/5 skeletal myoblasts damage induced by hydrogen peroxide in vitro and find an effective way to improve the survival of transplanted cells. Methods L6 SKMs were cultured and divided into control group(no treatment) , H202 group (24 hours of 1.00 mmol/L HE0: treatment) and DZ + H202 group(pretreated with 200 μmol/L DZ for 30 rain before 1.00 mmol/L H202 treatment for 24 hours). Effects of DZ pretreatment on L6 skeletal myoblasts damage by. oxidative stress were detected by HE staining, MTT method, LDH measurement, immunoeytoehemistry and flow cytometry. Results DZ pretreatment can improve the morphological structure of injured L6 skeletal myoblasts, decrease cell apoptosis, increase the survival of transplanted cells obviously. Conclusions DZ oretreatment has the obvious orotective effects on damaged L6 skeletal mvoblasts.
