摘 要:目的研究河南地区阴道毛滴虫临床分离株与人型支原体共生情况,并观察其共生对阴道毛滴虫AP33基因序列的影响。方法应用人型支原体及AP33基因的特异性引物对41株阴道毛滴虫临床分离株进行PCR扩增,扩增产物进行1.5%琼脂糖凝胶电泳检测,对AP33基因扩增产物测序,测序结果与GenBank已知序列比对,绘制进化树。结果有34株阴道毛滴虫扩增出人型支原体的特异性片段,大小为334bp;所有虫株均扩增出AP33基因特异性片段,碱基序列相似性为96.8%~99.3%,有多个位点发生突变。进化树分析提示人型支原体的共生对AP33基因序列有一定影响。结论河南地区阴道毛滴虫临床分离株与人型支原体共生具有普遍性,其共生对阴道毛滴虫的基因序列有一定影响。Objective To study the symbiosis of Mycoplasma hominis and Trichomonas vaginalis clinical isolates and observe their symbiotic effect on AP33 sequences. Methods Primers specific to M. hominis and the AP33 gene were used for PCR amplification of 41 T. vaginalis clinical isolates. Amplification products were analyzed using 1.5 % agarose gel electrophoresis. Amplification products of the AP33 gene were sequenced, and a phylogenetic tree was constructed by comparing sequencing results with the known sequences in GenBank. Results A 334-bp sequence specific to M. homi- nis was amplified from 34 of the 41 T. vaginalis clinical isolates. A sequence specific to the AP33 gene was amplified from all of the clinical isolates. The similarity of base sequences was 96.8%-99.3%, though there were mutations in several bases. Phylogenetic tree analysis suggested that there is a symbiotic effect of M. hominis on the AP33 gene se- quence to an extent. Conclusion The symbiosis of M. hominis and T. vaginalis clinical isolates is ubiquitous in Henan Province. This symbiosis has affected the gene sequences of T. vaginalis to an extent.
