摘 要:建立了测定忽地笑鳞茎中石蒜碱和加兰他敏含量的高效液相色谱分析方法,同时应用该方法测定了雪峰山不同海拔高度忽地笑居群鳞茎中石蒜碱和加兰他敏含量。色谱柱Agilent Eclipse XDB-C18(150 mm×4.6 mm,5μm),检测波长288 nm,流动相0.1%磷酸水溶液-甲醇(65:35),柱温25℃,流速1.0 ml/min,进样量20μL。研究结果表明,不同海拔忽地笑居群鳞茎中石蒜碱和加兰他敏含量存在一定差异,其中海拔678 m忽地笑居群鳞茎中石蒜碱和加兰他敏含量均最高,分别约为445.23μg/g和75.71μg/g,与其它海拔居群相比,差异显著。该方法精确、简便、灵敏度高。雪峰山地区海拔约678 m地带较适于以提取石蒜碱和加兰他敏为目的忽地笑栽培。A high performance liquid chromatographic (HPLC) method was developed to determine the contents of lycorine and galantamine in the bulb of Lycoris aurea, and analyze the contents of them in Lycoris aurea populations from different altitudes in Xuefeng Mountain. The chromatographic conditions was as following: 20 μL of samples or standards were injected into the Agilent E- clipse XDB - Cls ( 150 mm x 4.6 mm, 5 μm) column at 25 oC and eluted with mobile phase of water ( including 0.1% phosphoric acid) 65 : methanol 35 at flow rate of 1.0 mL/min, and the eluents were detected by absorption at 288 nm. The results showed that the contents of lycorine and galantamine of the populations from the different altitudes were different, among which, the contents of them of the population at altitude 678 m were all the highest, about 445.23 μg .g- 1 and 75.71 μg . g- 1 respectively. The difference was significant compared with the others. This method was simple, rapid and highly. It was more suitable for cultivating Lycoris au rea in the region of altitude about 678 m in order to extract lycorine and galantamine from the bulb.
