摘  要:从陕西省某大型养鸡场的疑似新城疫病鸡群中分离到1株新城疫病毒。对该新城疫病毒陕西分离株进行F基因的扩增、序列测定和遗传进化分析。结果显示,该新城疫病毒分离株的 F基因全长为1662 bp ,F蛋白裂解位点区的氨基酸序列为112R-R-Q-K-R-F117,具备强毒株的特征;同源性分析表明,该新城疫病毒瘾分离株与国内外流行的基因Ⅶ型强毒株之间的同源性较高,在92.4%~98.0%之间,其中与国内流行的基因Ⅶd亚型毒株的同源性最高,达97.3%以上,而与现用疫苗毒株LaSota(DQl95265)、F48E9(AY508514)、B1(AF309418)、V4(AF217084)、Mukteswar(EF201805)等的同源性较低,在83.2%~86.0%之间;系统进化树分析表明,该新城疫病毒分离株与2个国内流行的基因Ⅶ d亚型参考株 FJ882014和FJ608335处在同一个进化分支上,亲缘关系最近。A Newcastle disease virus (NDV ) was isolated from ND suspected chickens of a scale chicken farm in Shaanxi Province .The F gene of the Newcastle disease virus (NDV) Shaanxi isolate was amplified and genetic evolution analyzed .The results showed that the whole nucleotide sequence length of F gene was 1662bp .The sequence of amino acid at the cleavage site of F gene is 112R-R-Q-K-R-F117 ,which is consistent with the characteristics of strong NDV strain .Comparison of the NDV isolate with the strong NDV isolates of gene VII of epidemic strains from different countries showed that the nucleotide homology was 92 .4%-98 .0% ,and in which the comparison with the VIId were above 97 .3% .Comparison of the NDV isolate with the current vaccine strains such as Lasota (DQl95265 ) ,F48E9 (AY508514 ) ,B1 (AF309418) ,V4(AF217084) ,Mukteswar (EF201805) and so on showed that the nucleotide homology was 83 .2%-86 .0% . The phylogenetic tree analysis demonstrated that the NDV isolate and VIId strains of FJ882014-LN and FJ608335-XJ were located in the same cladogram branch .
