摘  要:目的寻找糖尿病肾病敏感的血浆分子标志物。方法对8例早期糖尿病肾病患者的血浆和正常血浆进行蛋白质组学荧光差异双向电泳(2-D DIGE)分析,选择表达差异大于1.5倍的蛋白斑点进行质谱分析。结果成功建立糖尿病肾病和正常血浆的胶内差异双向凝胶电泳图谱,通过分析共鉴定出13种差异蛋白质,包括C3补体、C4补体、载脂蛋白E等。结论 2-D DIGE能够客观地显示糖尿病肾病与正常血浆之间的蛋白质表达差异,本研究鉴定的13种蛋白有可能为糖尿病肾病的早期诊断提供潜在的血浆分子标志物。Objective To explore the sensitive blood plasma molecular markers in diabetic nephropathy(DN). Methods Two-di- mensional fluorescence difference gel eleetrophoresis (2D-DIGIt.) was used to analyze early DN patients plasma(n=8) and normal plasma, proteins that showed differential expression of a 1.5 fold change were analyzed by MALDI-TOF/TOF mass spectrometry. ResuRs 2D-DIGE maps of plasma proteins in patients with DN and normal plasma were established successfully. We validated 13 differentially expressed proteins detected by 2D-DIGE,including complement component C3. complement component C4.apolipopro- tein E,etc. Conclusion Proteomic analysis can objectively revealed the differences of protein expression between the two kinds of blood plasma. The 13 proteins might be the potential plasma molecular markers for the clinical diagnosis of diabetic nephropathy.
