摘 要:目的研究掌叶大黄(RheumpalmatumL.)的化学成分。方法采用反复柱硅胶色谱、制备薄层色谱、凝胶柱色谱(SephadexLH-20)、反相中低压柱色谱、制备高效液相色谱(PRE—HPLC)等手段进行分离纯化,根据理化性质及光谱数据确定化合物的结构。结果从掌叶大黄体积分数95%乙醇提取物的乙酸乙酯萃取物中分离得到6个化合物,分别鉴定为:2-乙酰基-3,5-二羟基苯乙酸甲酯(methyl2-acetyl-3,5-dihydroxyphenylacetate,1)、北美圣草素(eriodictyol,2)、二氢山萘酚(di—hydrokaempferol,3)、2-甲基-5-羧甲基-7-羟基色原酮(2-methyl-5.carboxymethyl-7.hydroxy-chromone,4)、2-甲基-5-甲氧羰甲基-7-羟基二氢色原酮(2-methyl-5-methoxycarbonylmethyl-7-hydroxychromanone,5)和2-甲基-5-甲氧羰甲基-7-羟基色原酮(2-methyl-5-methoxycarbonylmeth—yl-7-hydroxychromone,6)。结论化合物1—3为首次从大黄属植物中分离得到,化合物5为-新天然产物,化合物6为一新化合物。Objective To isolate and identify the chemical constituents from Rheum palmatum L. Methods The chemical constituents of R. palmatum L. were isolated and purified by multiple chromatographic tech- niques ( such as silica gel chromatography, Sephadex LH-20 and HPLC etc. ). These compounds were iden- tified by analysis of the spectral data and chemical properties. Results Six compounds were isolated from the ethyl acetate extract part of the R. palmatum L. ethanol extract and their structures were identified as methyl 2-acetyl-3,5-dihydroxyphenylacetate ( 1 ), eriodictyol ( 2 ), dihydrokaempferol ( 3 ), 2-methyl-5-carboxym- ethyl-7-hydroxychromone ( 4 ), 2-methyl-5-methylcarboxymethyl-7-hydroxychromanone ( 5 ), 2-methyl-5- methylcarboxymethyl-7-hydroxychromone (6). Conclusions Compounds 1-3 are isolated from Rheum ge- nus for the first time. Compound 5 is a new natural product. Compound 6 is a new compound.
