雷耐酸锶预防大鼠糖皮质激素性骨质疏松的实验研究

摘  要:目的研究雷耐酸锶对地塞米松诱发的继发性骨质疏松的预防作用。方法将80只SPF级SD大鼠,随机分为4组,正常组:常规喂饲;模型组:常规喂饲60天,第61天开始予地塞米松1 mg/(kg·d),90 d;预防Ⅰ组:连续60天给予雷耐酸锶75 mg/(kg·d),第61天开始,给予雷耐酸锶75 mg/(kg·d)和地塞米松1 mg/(kg·d),持续90 d;预防Ⅱ组:常规喂饲60天,从第61天开始,给予1 mg/(kg·d)地塞米松和75 mg/(kg·d)的雷耐酸锶,持续90天。实验结束,取股骨和第五腰椎进行骨密度检测,股骨进行骨形态计量学检测。结果预防组骨小梁面积百分数、骨小梁厚度及骨小梁数目均有改善,股骨周长百分数、骨形成率、成骨细胞数均提高,而破骨细胞数下降。预防Ⅰ组破骨细胞数少于预防Ⅱ组,而且预防Ⅰ组成骨细胞数高于预防Ⅱ组,两组间差异有统计学意义(P<0.05)。结论动物实验证明,雷耐酸锶对地塞米松诱发的糖皮质激素性骨质疏松有预防作用。Objective To investigate the preventive effect of strontium ranelate on dexamethasone induced osteoporosis. Methods 80 SD rats were randomly divided into 4 groups with 20 rats in each group. Normal group were feed by conventional. Model group were feed by con- ventional 60 days. Since the sixty- first day, the mouse were feed by dexamethasone 1 mg/( kg · d) for 90 days. The prevention group I: were feed strontium ranelate 75 mg/( kg ·d) for 60 days. Since the sixty - first day, the mouse were feed with strontium ranelate 75 mg/( kg · d), dexamethasone 1 mg/( kg ·d) , for 90 days. Prevention group II : were feed conventional 60 days. Since the sixty - first day, the mouse were feed strontium ranelate 75 mg/( kg · d) and dexamethasone 1 mg/( kg ·d) , for 90 days. At the end of the experiment, femur and fifth lumbar vertebra was detected by BMD. Femoral bone was used for histomorphometry detection. Results In the prevention group, percentage of trabecular area, trabecular thickness, trabeclar number percentage of femur perimeter, bone formation rate and the numbers of osteoblast were improved. However, osteoclast number decreased. The numbers of osteoblast in the prevention group I was higher than that in prevention group II. The osteoclast num- ber in the prevention group I was lower than that in prevention group II. There was significantly difference between two groups ( P 〈 0.05 ). Con- clusion The animal experimentation demonstrated that strontium ranelate may prevent glucocorticoid - induced osteoporosis.
