摘 要:为了探讨外源DNA导入诱导番茄变异与外源DNA浓度与处理温度的关系,采用浸胚法用不同浓度的大豆DNA溶液浸泡番茄种子,并置于不同温度下进行暗处理。对所获得当代(D1)植株叶片中过氧化物同工酶(POD)酶谱、酶活及叶片中可溶性蛋白含量等生理生化指标进行分析测定,结果表明:与受体对照组相比,D1代试验组的过氧化物同工酶酶谱有差异,在DNA浓度为341.25μg/mL、温度25℃处理下酶带增数最多且颜色最深;在DNA浓度为136.50μg/mL、温度20℃处理下POD的活性最强;在DNA浓度为341.25μg/mL、温度35℃处理下叶片中可溶性蛋白含量增加最多,增幅为1.79%。In order to discuss the effect of exogenous DNA concentration and temperature on introduction of tomato variation, the exogenous DNAs of soybean were introduced into the tomato by different concentration DNA solution by soaking embryo method in different temperature. The result showed that exogenous DNA introduction caused the change of the zymogram and activity of peroxidase isozyme (POD) and the soluble protein content in leaves of tomato. When the concentration of DNA was 341.25 p.g/mL and the temperature was 25℃, the number of enzyme band was the largest and the colour of enzyme band was the deepest as well. When the concentration of DNA was 136.50 Ixg/mL and the temperature was 20℃, the activity of the POD was the highest. When the concentration of DNA was 341.25 μg/mL and the temperature was 35℃, the soluble protein content addition was the most, increased by 1.79%.
