吸入麻醉药对大鼠心肌的保护作用

摘  要:目的观察吸入麻醉药对大鼠心肌缺血再灌注心肌的保护作用。方法健康雄性Wistar大鼠随机分成5组(各10只):假手术组、模型组及低、中、高实验组,建立大鼠心肌缺血再灌注模型,模型组的TNF-α、ICAM-1表达量均有所上升,ERK1/2表达量下降(P<0.05);实验组在灌注前15 min,分别吸入1%,2%,3%七氟烷。观察各组的肿瘤坏死因子(TNF-α)、细胞间粘附分子1(ICAM-1)及激活细胞外信号调节激酶1/2(ERK1/2)mRNA转录水平变化。结果与模型组对比,低、中和高实验组的TNF-α、ICAM-1表达量下降,而其ERK1/2表达量上升(P<0.05)。结论吸入麻醉药能有效地控制心肌TNF-α、ICAM-1及ERK1/2表达异常升高,对大鼠心肌具有一定的保护作用。Objective To observe protection effects of inhalation anes- thetics on the myocardial in rat with myocardial ischemia - reperfusion. Methods A total of 50 healthy male Wistar rats were randomly divided into 5 groups( 10 in each) : sham group, model group, low , middle and high tests groups. The myocardial ischemia reperfusion model of rats was established. The sham group only but not ligated the left anterior descending coronary artery, then three tests groups in the perfusion of inhalation anesthetics the first 15 min were exposed to 1% , 2% , 3% and seven halothane. The level changes of tumor necrosis factor ( 1/2TNF - α), extracellular signal -regulated kinase (p -ERK1/2), the transcription of mRNA intercellular adhesion molecule 1 ( ICAM - 1 ) express were observed. Results Compared with the model group, the TNF - ICAM - 1 expression decreased in three tests groups , while the ERK1/2 expression increased ( P 〈 0. 05 ). Conclusion Inhalation anesthetics can effective control the myocardial TNF- α, ICAM -1 and ERK1/2 expression, has a protective effect on rat myocardium.
