独活寄生汤水提物对软骨细胞G_1期调控因子mRNA表达的影响

摘　　要：目的：探讨独活寄生汤水提物对软骨细胞G1期的影响。方法：取4周龄雄性sD大鼠，建立软骨细胞体外培养体系，第3代软骨细胞进行干预，采用MTY法检测不同浓度的独活寄生汤水提物对软骨细胞活性的影响；lit-PCR检测空白组、独活寄生汤水提物低、中、高剂量组（100，200，400mg·L-1）软骨细胞CyclinD1，CDK4，CDK6及P21mRNA的表达。结果：MTF法检测显示，在一定的药物质量浓度范围内（50～800mg·L-1），软骨细胞活性显著上升（P〈0．01）；干预24h后，CyclinD1，CDK4及CDK6mRNA表达量各剂量组明显增加（P〈0．05），200mg·L-1表达量最大（P〈0．01）；P21mRNA表达量降低，且200mg·L-1抑制作用最明显（P〈0．01）。结论：独活寄生汤水提物通过影响软骨细胞G1期调控因子mRNA的表达，促进软骨细胞的增殖。Objective： To investigate the effect of water extracts from Duhuo Jisheng decoction on chondrocyte Gt phase. Method ： Chondrocytes were collected from four-week-old SD rats to establish the chondrocyte in vitro culture system. The third genera- tion of chondrocytes was intervened. MTT method was used to measure the effect of water extracts from different concentrations of Du- huo Jisheng decoction on chondrocyte activity. The expressions of Chondrocyte Cyclin D1, CDK4, CDK6 and P21 mRNA in the blank group and low, middle and high-dose groups （ 100, 200,400 mg · L-1 ） were detected by RT-PCR method. Result： The MTF assay showed that the chondrocyte activity significantly increased within specific drug concentrations （50-800 mg· L-1 ） （P 〈 0. 0l ） ; After the intervention for 24 h, the expressions of CyclinD1, CDK4 and CDK6 mRNA in all dose groups notably increased （P 〈 0. 05 ）, with the maximum expressions at the concentration of 200 mg·L-1 ; The expression of P21 mRNA decreased, particularly at the concentra- tion of 200 mg· L-1 （P 〈 0. 01 ）. Conclusion： Water extracts from Duhuo Jisheng decoction can promote chondrocyte proliferation by effecting the expression of chondrocyte G1 phase regulator mRNA.
