摘 要:选择生长速度不同的高邮鸭和金定鸭,对其胚胎期和出雏早期肝脏IGF-1 mRNA的表达规律及其与体质量的相关性进行了比较研究。采用实时荧光定量PCR方法研究鸭13、17、21、25、27胚龄和出雏后7日龄下肝脏胰岛素样生长因子1(IGF-1)mRNA的表达规律。试验结果证实:鸭肝脏IGF-1 mRNA在13胚龄已有表达。鸭早期发育过程中体质量和肝脏IGF-1 mRNA的表达水平均呈持续上升态势,高邮鸭13、21胚龄和7日龄肝脏IGF-1 mRNA的表达量极显著高于金定鸭(P<0.01),与日增体质量和绝对生长速率的变化情况相吻合。整个观察期内体质量变化和肝脏IGF-1 mRNA的表达量均呈现极显著的品种和时间特异性(P<0.01),并且受到品种和日龄交互作用的极显著影响(P<0.01)。从品种或性别的角度进行分析,均发现鸭发育早期肝脏IGF-1 mRNA的表达与体质量的变化趋势一致,且呈极显著正相关(P<0.01),提示肝脏IGF-1 mRNA的表达可能在鸭早期发育过程中发挥着重要作用。The present experiment was conducted to study the tissue expression characterization of IGF-1 mRNA during embryogenesis and post-hatching development in different breeds of ducks. We have compared the ontogeny of body weight and expression of insulin- like growth factor-1 (IGF-1)mRNA in liver at the embryonic day 13,17,21,25,27 and the age of 7 day of Gaoyou duck and Jinding duck. At the first time ,the trial presented that duck liver IGF-1 mRNA has been detectable at day 13. During early development, the body weight and hepatic 1GF-1 mRNA expression both presented a rising tendency persistently, and the hepatic IGF-1 mRNA expression of Gaoyou duck group at 13,21 embryonic day and 7 day were hi^gher than that of Jiuding duck group (P〈0.01), which matched the changes of the daily gain and the relative growth rate of the two breeders. During the experimental period, extremely significant variety( P〈O. 01 ) and time( P〈0.01 ) specificity was found in developmental change of body weight and hepatic IGF-I mRNA expression,in which the significant interaction effect existed. Analyzed regardless of breeder or gender differences ,the profiles of IGF-1 mRNA levels were compared to developmental changes in body weight, and the results showed a progressive increase in hepatic IGF-1 mRNA which continued after the birds body weight,and both appear to show a strong positive linear relationship in duck(P〈0.01 ). It was suggested that the differential expression of hepatic IGF-1 mRNA may be involved in the early growth of ducks.
