秋茄ERF抗逆转录因子的克隆及其功能分析

摘　　要：利用RACE(Rapid-Amplification of cDNA Ends)技术,以深圳红树林的秋茄(Kandelia obovata)作为材料,克隆一个ERF类转录因子基因cDNA全长序列,命名为KcERF(Genebank,序列号:GU593721)。序列分析结果表明,该基因完整开放阅读框873bp,编码290个氨基酸。比对结果显示,有一个典型的AP2结构域,空间结构具有1个α螺旋和3个β折叠,属于AP2/EREBP家族中的ERF类转录因子。KcERF亚细胞定位在细胞核中,其编码蛋白在酵母中有转录激活活性。此外,KcERF受到高盐胁迫后,转基因型烟草(Nicotiana tabacum)的株高和叶面积、含水量、MDA含量、可溶性糖含量、电导率均表现出对盐的耐受作用,暗示本研究结果可以为转基因耐盐棉花(Gossypiumspp.)和牧草的培育提供新的基因来源。Taking Kandelia obovata in Shenzhen mangrove as material, the full length eDNA sequence of a transcription faetor gene named as KcERF（GenBank, Accession No. ：GU593721） was cloned, using RACE（Rapid Amplification of cDNA Ends） technology, in this study. Results of sequence analysis showed that, the full-length of this gene is 873 bp, and encodes a protein containing 290 amino acids. The alignment results showed that this protein contains a cope of typical APE DNA binding domain, and a a-he lix and three 13-pleated sheets in space structure. So this gene is a ERF transcription factor o{ AP2/ EREBP. Subcellular localization analysis manifested that KcERF is located in the nucleus, and encodes protein which has transcriptional activity in yeast. In addition, transgenic Nicotlanatabacum, which con rains KcERF, performed tolerance to salt in all aspects, including plant height, leaf area, moisture con tent, MDA content, soluble sugar content, electrical conductivity and so on. And this implied that this study can provide new gene source for transgenic xerophytic Gossypium spp and forages.
