强精煎对少弱精子症模型大鼠超微结构的影响

摘　　要：目的探索强精煎对少弱精子症模型大鼠睾丸附睾及精子超微结构的影响。方法 80只雄性成年SD大鼠随机分成4组。正常组大鼠每天给予生理盐水;模型组予奥硝唑(ORN)800 mg/(kg·d);黄精赞育胶囊组予[ORN 800 mg/(kg·d)+黄精赞育胶囊400 mg/(kg·d)];强精煎组给予[ORN 800 mg/(kg·d)+强精煎配方颗粒10 g/(kg·d)]。灌胃4周后取睾丸及附睾组织,透射电镜观察生精上皮、附睾上皮及精子的细胞与亚细胞结构,同时进行常规病理观察。结果模型组附睾管内仅见少量精子,生精细胞核膜崩解、细胞器模糊、缺失,精子线粒体鞘部分缺失或完全崩解碎裂。强精煎组附睾管腔内可见大量精子,生精细胞核清晰,线粒体等细胞器丰富,结构清晰,精子线粒体鞘基本完整,微管9+2结构及致密纤维清晰。结论强精煎中药可明显修复大剂量奥硝唑导致的睾丸附睾及精子超微结构损伤,恢复并维持一定程度的生精功能,可能是其治疗少弱精子症的机制之一。Objective To observe the affects of Qiangjing Decoction on the ultrastructure of testis epididymis and sperms in rat with oligoasthenospermia. Methods Randomly divide 80 adult SD rats into normal group, model group, Qiangjing Decoction group and HuangJingzanyu capsule group（ n = 20）. Delivery method：normal group：normal saline given daily;model group：ORN （ 800 mg/kg·d） ; HuangJingzanyu capsule group： ORN （ 800 mg/kg ~ d ） ＋ Huang Jingzanyu capsule （ 400mg/kg· d ） ; Qiangjing Decoction group：ORN （800 mg/kg · d ） ＋ Qiangjing Decoction（ 10 g/kg ·d）. Four weeks after the administration, we take the testis and epididymis of the rats to run a transmission electron microscopy test, so to observe the cellular and subcellular structures of the ratsgeminiferous epithelium, epididymal epithelium and sperm. We also take Routine pathological observation on them. Results Model group：only a small amount of sperms can be seen in the epididymal duct;the nuclear membranes of the spermatogenic cells are disintegrated ; the organelles of these ceils are fuzzy or missing ; the mitochondrial sheath of the sperms are partially missing or completely disintegrated. Qiangjing Decoction group ： a large amount of sperms can be seen in the epididymal duct, the spermatogenic nuclei can be seen clearly, the organelles are abundant, the structures of organelles are seen clearly ; the mitochondrial sheath of the sperms are basically complete, the 9 ＋ 2 structure and the dense fibers are clear. Conclusion Qiangjing Decoction can fix the ultrastructural damage of the testis and epididymis caused by ornidazole, to restore and maintain normal spermatogene- sis to a certain degree. This may be one of the mechanisms of Qiangjing Decoction healing the oligoasthenospermia.
