资冲颗粒体外上调人脐带静脉内皮细胞的VEGFR-2等8种基因转录水平的研究  被引量:2

摘  要:目的研究一种中药专利药方资冲颗粒促进更年期妇女血管生成的机制。方法选用HUVEC细胞作为体外模型;制备资冲颗粒的大鼠含药血清:使用含药血清干预HUVEC;对干预后的细胞进行以下两方面检测:使用含药血清连续干预HUVEC 5 d,期间每天检测细胞形态、培养上清液中雌二醇(E2)水平变化和被干预的HUVEC的生长速率;使用含药血清干预3 d,而后用实时荧光定量PCR(Qpcr)检测8个血管生成相关基因的转录表达水平。结果资冲颗粒含药血清不明显改变HUVEC的细胞形态、培养上清液中的E2水平变化和HUVEC的生长速率;资冲颗粒含药血清明显上调了VEGFR-2、VEGFR-1、ANGPT-1、TIE-2和ER-1等基因的转录表达水平,同时明显下调VEGF-A基因的转录表达水平,而ER-2和ANGPT-2基因的转录表达水平无明显改变。结论研究初步阐明了资冲颗粒促进人血管生成的机制。Objective To investigate the mechanism of Zi Chong Granule ( ZCG), a patented herbal formula in Traditional Chinese Medicine (TCM) ,to promote angiogenesis in women in menopause. Methods HUVEC were chosen as the in vitro model;the ZCG drug serum was prepared ; HUVEC were treated with the drug serum ; the next investigation has two parts : ( 1 ) HUVEC were treated for 5 days and during the period, the morphology, estradiol ( E2 ) level in the culturing medium and the growth rates of the treated HUVEC were analyzed ; (2) HUVEC were treated for 3 days and after that, the transcriptional expression of 8 angiogenesis - related genes were analyzed by real - time fluorescence quantitative PCR (Qpcr). Results ( 1 ) The ZCG drug serum did not significantly change the morphology, E2 level in the culturing medium and the growth rates of the treated HUVEC ; ( 2 ) the ZCG drug serum can significantly up -regulate the transcription of VEGFR- 2, VEGFR- 1, ANGFT- 1 ,TIE -2, ER- 1, and that of VEGF - A were down - regulated, meanwhile, that of ER - 2 and ANGPT - 2 did not change significantly. Conclusion This re- search preliminarily clarified the mechanism of ZCG to promote angiogenesis in truman.
