摘  要:目的研究怀牛膝中总皂苷成分的提取、纯化工艺,确定牛膝总皂苷类成分体内治疗痛风性关节炎疗效。方法以齐墩果酸为指标,采用单因素考察及正交实验设计确立牛膝总皂苷最佳提取工艺,利用大孔吸附树脂法对牛膝有效成分进行纯化工艺研究。以大鼠足肿胀为指标,对纯化后的牛膝总皂苷治疗痛风性关节炎的作用进行评价。结果牛膝总皂苷最佳提取工艺为以70%乙醇作为溶剂,提取两次,每次提取1 h,加溶剂量12倍。最佳纯化工艺为D101型大孔吸附树脂装柱,径高比约为1∶5,树脂比生药量为1∶0.2上样,上样流速为1.5 BV·h-1,4 BV去离子水和50%乙醇洗脱,洗脱速度为1.5 BV·h-1,50%乙醇洗脱液洗脱部分为牛膝总皂苷。牛膝总皂苷提取物灌胃给药(生药剂量8 g·kg-1),1次/d,连续给药3 d,可明显缓解由弗氏完全佐剂引起的大鼠足趾肿胀。结论牛膝总皂苷经本工艺提取、纯化后,其含量可达55%以上,简单易行,适合工业化生产,牛膝总皂苷对大鼠佐剂性关节炎具有抑制作用,为进一步研究有效成分治疗湿热痹证提供前期试验基础。Objective To optimize the extraction and purification technology of total saponins from Achyranthes bidentata Blume for damp arthralgia. Methods The extraction and purification conditions were examined by using oleanolic acid as main indices. The effect of total saponins ( 8 g · kg - 1 ) on plantar swelling was examined after the total saponins was given into the rats of damp arthralgia induced by Freund's complete adjuvant. Results The optimized extractions could be obtained by adding 12 times amount of 70% ethanol for twice, 1 hour each time. The best purification conditions were specified as follows:using D101 macroporous res- in, the sampling ratio of resin weight to raw material was 1 : 0.2 with a flow rate of 1.5 BV · h - 1 and 4 BV 50% aqueous - etha- nol as elusion solvent. Total saponins of A. bidentata Blume. had obvious effect on plantar swellings. Conclusion The total saponins were the active components for damp arthralgia. And the total saponins content in the extract could reach above 55% , and this method is simple and practical
