摘 要:目的观察壮骨镇痛胶囊对裸鼠乳腺癌移植瘤黏附迁移相关指标的影响,探讨其抑制乳腺癌转移的可能机制。方法各组裸鼠接种乳腺癌制备成移植瘤模型,随机分为空白模型组、预防组、治疗组和阳性对照组,按照给药方案用药,计算各组抑瘤率并进行比较;免疫组化方法检测各组标本黏附斑激酶FAK、磷酯酰肌醇3-激酶PI3-K及丝/苏氨酸蛋白激酶MEK1/2的蛋白表达活性,比较其组间差异。结果各用药组裸鼠体重生长情况及移植瘤体积与模型组相比较均存在差异,差异均具有统计学意义(P<0.05);抑瘤率分别为预防组56.84%、治疗组36.49%、阳性对照组35.45%。模型组与各灌胃组比较,FAK、PI-3K及MEK1/2表达活性较高,差异具有统计学意义(P<0.01),壮骨镇痛胶囊预防组与参一胶囊阳性模型对照组比较,差异无统计学意义(P>0.05),其余各组差异均有统计学意义(P<0.05)。结论壮骨镇痛胶囊可通过抑制黏附斑激酶FAK蛋白的表达,诱导下调乳腺癌移植瘤组织磷酯酰肌醇3-激酶PI3-K及丝/苏氨酸蛋白激酶MEK1/2的蛋白表达活性,抑制癌细胞的黏附迁移。Objective To study the probable mechanism of action that ZZC (Zhuanggu Zhentong capsules) inhibit the migrations of breast cancer. Methods Implanted tumors were established and were randomly divided into 4 groups at first, i. e. modeling group (MG), preventing group (PG) with ZZC, treating group (TG) with ZZC and positive controlling group (PCG) treated by Shenyi capsules( SC), and treated with various treating procedures. We evaluated the inhibiting rate of tumor growing by light microscopy respectively ;immunohistochemieal ABC procedures were taken to determine the expressive activities of protein system in the terms of biodynamic correlation, such as FAK, PI3 - K and MEK1/2, among different groups with a comparative analysis. Results There were statistically significant differences in the volumes of tumors among the nude mice of PG,TG and PCG ( P 〈 0.05 ) ,with the inhibitory rates of tumor growing being 56.84% ,36.49% and 35.45% for them respectively. As determined by SABC immunohis- tochemistry followed by image analysis on the cellular adhesion - relating proteins in sections of various groups, the expressive activities of FAK,PI3 - K and MEK1/2 were much higher in the samples of MG than that of PG,TG and PCG,with significantly statistical differences among them (P 〈0. 01 ). Conclusion It can be concluded from this experimental study that ZZC bring about inhibitory effect on the expression inhibition of FAK to induce decreased expressive activity of PI3 - K and MEK1/2 in these cells so as to destroy the formation of fibrous skeletons in an inhibited way and bring about through its intervening effect on their biodynamie mechanism.
