摘 要:目的研究提取甘草酸后的甘草废渣中的有效成分—异甘草素、光甘草定对酪氨酸酶活性抑制的影响。方法以L-酪氨酸作为底物,采用比色法测定异甘草素、光甘草定对酪氨酸酶活性的影响,并以Lineweaver-Burk双倒数法绘制酶动力学曲线,推断抑制类型。结果异甘草素、光甘草定都不同程度的抑制酪氨酸酶活性,其中异甘草素抑制酪氨酸酶单酚酶的IC50值为1.66 mmol·L-1,对二酚酶无抑制效果;光甘草定抑制酪氨酸酶单酚酶、二酚酶的IC50值分别为31.16μmol·L-1、20.38μmol·L-1;且光甘草定对酪氨酸酶的效应表现为竞争型抑制。结论该研究可为开发利用废弃甘草渣中有效成分作为植物源美白添加剂提供依据。Objective To study the inhibitory effects of active constituents of isoliquiritigenin and glabridin which exact from Glycyrrhiza waste residue on the activity of tyrosinase. Methods The inhibition of isoliquiritigenin and glabridin on tyrosinase were estimated with the colorimetry method using the L - Tryosine as the substrate and its inhibitory pattern was determined according to the Lineweaver - Burk curve. Results Isoliquiritigenin and glabridin could inhibit the activity of tyrosinase in defferent degrees, the IC50 values of Inhibition of tyrosinase monophenolase by isoliquiritigenin were 1.66 mmol·L^-1, have no inhibitory effects of tyrosinase diphenolase ;The ICs0 values of Inhibition of tyrosinase monophenolase and diphenolase by glabridin were 31.16 μmol·L^-1 and 20.38μmol·L^-1, and its inhibitory mechanism belonged to be competitive. Conclusion This study has provided the evidence for active constituents from Glycyrrhiza waste residue could be considered as a botanical whitening agent.
