摘 要:目的通过观察隔药灸命门穴对痛经模型大鼠血清雌激素(Estrogen,E2)和孕激素(Progesterone,P)水平的影响,探讨隔药灸命门穴治疗原发性痛经的作用机制。方法将100只雌性SD大鼠随机分为造模组(80只)和空白组(20只),以缩宫素建立大鼠原发性痛经模型。空白组为正常对照组(A组),并将造模成功的80只大鼠随机分为隔药灸组(B组)、针刺组(C组)、药物组(D组)、模型组(E组)。比较各组大鼠的扭体反应潜伏期和30 min内扭体数并用放免法检测各组大鼠血清E2和P的水平。结果①空白对照组无扭体,模型组扭体次数较多,且有扭体潜伏期,两组比较有非常显著性意义(P<0.01)。与模型组相比,隔药灸组、针刺组以及药物组均能明显延长首次扭体潜伏期,且能减少30 min内扭体反应次数(P<0.01)。隔药灸组、针刺组以及药物组扭体潜伏期比较,差异无统计学意义(P>0.05),但与药物组相比,隔药灸组及针刺组均能明显减少30 min内扭体反应次数(P<0.01);且隔药灸组与针刺组相比,隔药灸组30 min内扭体次数最少(P<0.01)。而且三组比较,隔药灸组扭体抑制率最高。②与空白对照组比较,隔药灸组、针刺组、药物组及模型组大鼠血清E2含量显著增加,P含量显著降低(P<0.01);与模型组比较,隔药灸组、针刺组及药物组E2显著降低,P显著增加(P<0.01);与药物组比较,隔药灸组及针刺组E2显著降低,P显著增加(P<0.01);与针刺组比较,隔药灸组E2显著降低,P显著增加(P<0.01)。结论隔药灸治疗原发性痛经可能与降低痛经大鼠血清E2,增加血清P水平有关。Objective To observe the effect of drugpaste interposed moxibustion on the expression of estrogen( E2 ) and progestogen (P) in rats with primary dysmenorrheal, and in order to investigate the mechanism of drugpaste interposed moxibustion on Mingmen(DUd) in treating primary dysmenorrhea. Methods 100 female rats were randomized into Model Group(80 rats) and Control Group (20 rats), and primary dysmenorrhea model rats were established by extractum leonuri inspissatum(ELI). The blank control group( A Group), and 80 female model rats were divided into four groups, drug -paste interposed moxibustion group (B Group), acupuncture group( C Group), medicine group( D Group), model group( E Group). The latency period and the incidence rate of writhing body within 30 minutes were observed in each group and the serum levels of E2 and P were detected by RIA. Results (1)The latency period of writhing body was shorter and the incidence rate of writhing body was more in model group, com- pared with those of A Group(P 〈0.01 ). The latency period of writhing body was longer and the incidence rate of writhing body was less in B Group,C Group and D Group,compared with those of E Group(P 〈0. O1 ). There was no significant difference between B Group, C Group and D Group in the latency period of writhing body ( P 〉 0.05 ). But compared with D Group, the incidence rate of writhing body was less in C Group( P 〈0.01 ) and it was least in B Group( P 〈 0.01 ). Compared with B ,C and D Group, the control rate of writhing body was highest in B Group. (~)Compared with A Group, the serum levels of E2 in B, C, D and E Group were obviously ascend( P 〈 0.01 ), while the serum level of P was obviously decressed ( P 〈 0.01 ). Compared with E Group, the serum levels of E2 in B, C and D Group were obviously decressed( P 〈 0.01 ), while the serum level of P was obviously increased( P 〈0.01 ).
