摘 要:目的:观察CD166和TSPAN-1基因在结直肠癌组织中的表达及与临床病理因素的关系。方法:采用免疫组化SP法检测CD166和TSPAN-1蛋白在120例结直肠癌组织、20例结肠腺瘤组织及40例癌旁正常结直肠黏膜组织中的表达。结果:CD166蛋白在正常黏膜、结直肠腺瘤、结直肠癌中阳性率分别为0、20%、55%,组间差异具有统计学意义(P<0.01);TSPAN-1蛋白在正常黏膜、结直肠腺瘤、结直肠癌组织中阳性率分别为5%、20%、92%,组间差异均具有统计学意义(P<0.05);CD166和TSPAN-1蛋白的表达与结直肠癌的组织学分级及TNM分期有关(P<0.01);CD166和TSPAN-1在结直肠癌组织中的表达强度呈正相关(r=0.524,P<0.001),二者表达完全一致率高达36.7%(44/120)。结论:CD166和TSPAN-1基因的高表达与结直肠癌的侵袭和发展有关,两者在结直肠癌的细胞增殖、浸润和转移中具有协同作用。联合检测两者的表达对于进一步理解结直肠癌的生物学行为和判断预后有一定价值。Objective:To study the expression of CD166 and TSPAN - 1 protein in colorectal carcinoma, and investigate the relationship between the expression and clinicopathologic parameters of colorectal carcinoma. Methods:Immunohistochemical method was used to detect the expression of CD166 and TSPAN - 1 protein in 120 cases of colorectal carcinoma,20 cases of eoloreetal adenoma and 40 cases of normal colorectal tissues. Results: The positive expression rate of CD166 and TSPAN - 1 in colorectal carcinoma was significantly higher than that in coloreetal adenoma and normal colorectal tissues. And there was significant difference in expression of every group ( P 〈 0.01 ). The positive expression rate of CD166 and TSPAN - 1 was 55% and 92% in group colorectal carcinoma. Both 20% in group colorectal adenoma and 0% and 5% in group normal coloreetal tissues (P 〈0. 01 ). The expression of CD166 and TSPAN - 1 was positively related with histopathological grading and TNM stage (P 〈 0. 01 ). The expression of CD166 was positively related with the expression of TSPAN - 1 in colorectal carcinoma ( r = 0. 524, P 〈 0. 001 ). Conclusions: The over expression of CD166 and TSPAN - 1 gene are closely related to the invasion and growth of colorectal carcinoma. CD166 and TSPAN - 1 have synergistic effect on invasion and metastasis of malignant cells. Detecting the expression of both CD166 and TSPAN - 1 may be valueful in further understanding the biological behavior and predicting the prognosis of colorectal carcinoma.
