摘  要:目的 :利用小鼠耳廓ACD模型进行养血消风饮的药效学研究,从Th1/Th2网络平衡的角度进行其免疫学机制探讨,为研究药物的作用机理提供依据。方法 :用二硝基氯苯(DNCB)建立小鼠耳廓慢性ACD模型,随机分为4组:空白组、模型组、中药组、氯雷他定组,对比观察小鼠血清中IFN-γ和IL-4含量,以及小鼠病变耳组织中IFN-γmRNA及IL-4mRNA的表达情况,并进行相关性分析。结果 :养血消风饮减少模型小鼠血清中IFN-γ的含量和增加IL-4含量,并能促使小鼠病变耳组织中IFN-γmRNA低表达和IL-4mRNA的高表达。结论 :养血消风饮可能在转录水平和分子水平对ACD小鼠的Th1/Th2网络平衡失调起到调节作用。Objective : To test the efficiency of the herbal medicine Yangxue Xiaofeng Drink by using the mice models of ACD on its auricles. From the immune balance of Thl/Th2 to explore the immunological inhibition mechanism further in-depth. Methods: Chronic ACD mice model was built up by stimulating its auricles by DNCB, then all the mice were separated into four groups: blank model group, model group, traditional Chinese medicine treatment group, Western medicine control group. The levels of IFN-γ and IL-4 in the serum of ACD mice model were measured by using ELISA method, using RT-PCR to detect lesions local IFN- γ/mRNA, IL-4 mRNA expression, and then the correlation of serum cytokine protein expression and mRNA expression in lesions were analyzed. Results : Yangxue Xiaofeng Drink can raise level of IL-4 and reduce that of IFN- γ in serum of ACD model, while increase expression level of IL-4 mRNA and reduce that of IFN- γ mRNA in ear tissue. Conclusion : Yangxue Xiaofeng Drink may adjust the network of Th 1/Th2 immune balance of chronic ACD mice model at the transcriptional level and molecular level.
