摘 要:目的:建立去卵巢豚鼠脂代谢紊乱模型,探讨苦菜总黄酮对去卵巢豚鼠脂代谢的影响。方法:采用去卵巢方法建立豚鼠脂代谢紊乱模型,30只雌鼠,随机分5组,每组6只:①正常对照组;②模型组;③阳性药物对照组(戊酸雌二醇,80mg/kg·d);④低剂量组(苦菜总黄酮,0.1 mg/kg);⑤高剂量组(苦菜总黄酮,0.3mg/kg)。术后1周内所有去除卵巢豚鼠均连续阴道涂片检查。术后1周建模成功后,阳性药物对照组用戊酸雌二醇水溶液灌胃(80 mg/kg·d),苦菜总黄酮低剂量、高剂量组分别按0.1、0.3 mg/kg苦菜总黄酮液剂量灌胃,正常对照组、模型组每天等体积生理盐水灌胃,各组均1次/天,连续6周给药。术后每周称体质量,量体长,计算Lee's指数。实验结束后,检测各组豚鼠血清HDL、LDL、TG、CHO的浓度。结果:与模型组相比,苦菜总黄酮可降低去卵巢豚鼠Lee's指数,从第3周开始明显,其中高剂量效果好。与模型组比较,苦菜总黄酮各给药组豚鼠血清HDL含量均升高;LDL、TG、CHO含量则均下降(P<0.05或P<0.01),并呈一定的量效关系,以高剂量效果好。结论:苦菜总黄酮对去卵巢成年动物的脂代谢失衡有调节作用,显示类雌激素样功效,可通过调节血脂水平而预防绝经后妇女因肥胖导致心血管疾病的可能。Objective: To establish lipid metabolism disorder model of ovariectomized guinea pigs, explore the effect of sonchus ol- eraceus on lipid metabolism of ovariectomized guinea pigs. Methods : Ovariectomized method was used to establish lipid metabolism disorder model of guinea pigs, 30 female guinea pigs were randomly divided into five groups, six guinea pigs in each group: normal control group, model group, positive drug control group ( estradiol valerate, 80 mg · kg-1 ·d -1 ), low - dose group ( sonchus oleraceus, 0. 1 mg/kg), and high - dose group ( sonchus oleraceus, 0. 3 mg/kg) ; vaginal smear examination was conducted continuously within one week after sur- gery in the five groups; at one week after establishing model successfully, the guinea pigs in positive drug control group, low -dose group, and high - dose group were treated with corresponding drugs by intragastric administration, while the guinea pigs in normal control group and model group were treated with intragastric administration of isovolumetric normal saline, once a day for six weeks; body weight and body height data were obtained every week after surgery, Lee's index was calculated; after the experiment, the concentrations of serum high density lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TG), and total cholesterol (CHO) in the five groups were detected. Re- suits: Compared with model group, sonchus oleraceus reduced Leeg index, which was significant from the third week, especially in high - dose group ; compared with model group, serum levels of HDL in low - dose group and high - dose group increased, while the serum levels of LDL, TG, and CHO decreased, there were statistically significant differences ( P 〈 0. 05 or P 〈 0. 01 ), showing a certain dose - effect rela- tionship, especially in high - dose group. Conclusion : Sonchus oieraceus can regulate lipid metabolism disorder model of adult ovariecto- mized guinea pigs, which shows estrogen - like effect and can prevent the
