摘  要:目的研究磺酸化甲壳素和磺酸化壳聚糖的理化性质及其生物安全性,探讨其抗凝效果及对癌细胞生长的抑制作用。方法通过甲壳素/壳聚糖的羟基(尤其是C6-OH)与磺化剂(氯磺酸/浓硫酸混酸)发生酯化反应合成磺酸化甲壳素(S-CT)/磺酸化壳聚糖(S-CTS),并对S-CT和S-CTS的生物安全性进行评价;通过全血凝血实验对其抗凝效果进行初步研究,MTT比色法评价其对人宫颈癌细胞HeLa和人喉表皮样癌细胞HEp-2生长的影响。结论S-CT和S-CTS的生物安全性符合国家标准,具有显著的抗凝血效果,对HeLa和HEp-2两种肿瘤细胞的生长均有明显抑制作用,其中以1000μg/mL S-CTS抑制效果为最佳(P<0.05)。为甲壳素、壳聚糖磺酸化产物的生物学应用提供一定实验依据。Objective To study the physicochemical and biological properties of sulfonated chitin and sul-fonated chitosan, and explore its anticoagulant effect and its impact on cancer cell growth inhibilion. Methods Esterification reaction sutfonated chitin (S-CT)/sulfonated chitosan (S-CTS) occured by Chi-tin/Chitosan's hydroxy (especially the C6-OH), with a sulfonating agent (chlorosulfonic acid, eoncen trated sulfuric acid). The biological safeties of S-CT and S-CTS were evaluated. Preliminary studies of S-CTs and S-CTSs anticoagulant effect were done through the whole blood clotting experiments. The MTT eolorimetric method was used for the evaluation of its impact on cell growth of human cervical carcinoma HeLa cells and human laryngeal epidermoid carcinoma HEp-2 cells. Results SCT and S-CTS had good biological safeties as well as significant anticoagulant effect. They inhibited the growth of He-l.a cells and HEp 2 cells. The S-CTS(1000μg/mL)'s inhibitory effect was the best (P〈0. 05). This study provides some experimental evidence for biological applicalions of chitin's and chitosan's sulfon- ated products.
