摘  要:目的:探讨qRT-PCR检测中β-Actin Ct值与检测因子Ct值检出率和相关性分析。方法:以32只成年SD大鼠脊髓为实验样本进行两次qRT-PCR检测,并分别测β-actin Ct值和PENK、CNTF、PDGF Ct值,随后进行独立样本T检验、卡方检验、一元线性回归分析。结果:两次qRT-PCR检测β-Actin Ct值无统计学差异(P>0.05);两次检测均显示β-actin Ct值小于20与β-actin Ct值大于20对其他因子的检出率有显著性影响(P<0.05);β-actin Ct值与检测因子Ct值有显著的线性相关性,线性回归方程为y=-0.983+0.708x(y:欲检测基因Ct值,x:实际测得β-actin Ct值)。结论:qRT-PCR检测中可通过β-Actin Ct值预测需检测因子的Ct值。Objective: To investigate the relationship between β -Actin Ct value and the detection factor Ct value in qRT-PCR. Methods: spinal cord samples of 32 adult SD rats were employed in the qRT-PCR detection, and we measured β -actin Ct values and PENK Ct value, CNTF Ct value, PDGF Ct value independently, followed by T-test, chi-square test, one yuan linear regression analysis. Results: the first and the second qRT-PCR detection of β -Actin Ct value was no significant difference (P〉0.05); two tests have shown that β -actin Ct value of less than 20 and β -actin Ct values more than 20 was a key point of check rate for other factors (P〈0.05); β -actin Ct value and detection factors' Ct values have a significant linear correlation, the linear regression equation was y = -0.983 +0.708 x (y: for detection of gene Ct value, x: the actually measured value of β -actin Ct). Conclusion: via β -Actin Ct value prediction other factors Ct values in qRT-PCR detection.
