芍药舒筋片对C_(57)黑鼠膝骨关节炎的影响及其抗炎、镇痛作用研究

摘　　要：目的评价芍药舒筋片保护C57黑鼠软骨组织及其抗炎、镇痛作用,为其临床应用提供实验依据。方法将70只黑鼠随机分为空白对照组,模型对照组,阳性对照组,芍药舒筋片高、中、低剂量组。通过跑步机运动负荷成模后,灌胃给药4周,然后取膝关节进行病理形态学观察。采用小鼠耳郭肿胀、大鼠足跖肿胀模型,分为空白对照组,阳性对照组,芍药舒筋片高、中、低剂量组,分别干预5天后观察芍药舒筋片的抗炎作用;采用小鼠热板法和扭体法,分为空白对照组,阳性对照组,芍药舒筋片高、中、低剂量组,分别采取相应的干预措施,研究芍药舒筋片的镇痛作用。结果芍药舒筋片可明显减轻C57黑鼠膝骨关节炎关节软骨损伤程度;Mankin评分提示,阳性对照和治疗药组与模型组比较差异均有统计学意义(P<0.01),且芍药舒筋片高、中剂量组作用优于阳性药维固力组。芍药舒筋片能降低巴豆油致小鼠外耳郭的肿胀度和肿胀率,阳性对照组和给药组与空白组比较差异均有统计学意义(P<0.05,P<0.01);对鸡蛋清致大鼠足跖的肿胀度和肿胀率亦有明显的减轻作用,阳性对照组和高、中剂量给药组与空白组比较差异均有统计学意义(P<0.05,P<0.01);阳性对照组和高、中剂量给药组与空白组小鼠痛阈值比较,差异均有统计学意义(P<0.01);阳性对照组和给药组与空白组小鼠扭体次数比较,差异均有统计学意义(P<0.01)。结论芍药舒筋片可一定程度延缓运动负荷导致的C57黑鼠膝骨关节炎关节软骨退变,且有较好的抗炎镇痛作用。Objective To evaluate the protective effects of ＂Shaoyao Shujin Tablets＂ in Cs7 black mice cartilage tissue, and its anti-inflammation and analgesia to provide experimental basis for their clinical application. Methods Seventy black mice were randomly divided into 6 groups： blank control group, model control group, positive control group and ＇＂ Shaoyao Sbujin Tablets＂ groups （low, medium and high dose）, and modeled by treadmill exercise with load. Then knee joints were taken for pathological obsmwation after intragastrie administration for 4 weeks. Mice auricle and rats paw swelling models were divided into 5 groups to research its anti-inflammatory action after 5 days intervention. Mice were divided into blank control group, positive control group and ＂ Shaoyao Shujin Tablets＂ groups （ low, medium and high dose） by hot plate and twisting body methods. Corresponding treatments were used for different groups and its analgesic effect was studied. Results Histopathology observation showed that modeling by exercise with load was successful and ＂Shaoyan Shujin Tablets＂ could significantly reduce articular cartilage injury of knee osteoarthritis in C57 black mice. Mankin score indicated that positive and treatment groups both had significant differences statistically compared with the model group （ P 〈 0.01 ）, and the tablets in high, medium dose group were superior to positive drug ＂ Glueosamine Sulfate Capsules＂. ＂Sbaoyao Shujin Tablets＂ could reduce mice auricle swelling and their rates induced by croton oil and the positive and treatment groups both had significant differences statistically compared with the control group （ P 〈 0.05, P 〈 0.01 ）. Rats paw swelling and their rates induced by albumin were also reduced obviously and all the positive, high dose and medium dose groups had significant differences statistically compared with the control group （ P 〈 0.05 or P 〈 0.01 ）.
