龙蝎消水膏对艾氏腹水瘤小鼠VEGFmRNA、MMP-2mRNA、FLt-1mRNA表达的影响

摘　　要：目的探讨龙蝎消水膏对艾氏腹水瘤(EAC)小鼠的作用及其可能机制。方法 Balb/c-nu/nu裸鼠除正常组10只外,另外40只小鼠腹腔内注射0.2ml EAC细胞株悬液(瘤细胞浓度1×107/ml)制备腹水瘤小鼠模型。造模成功后随机分为4组,即对照组(予0.9%NaCl溶液)、中药外敷组(予龙蝎消水膏组)、西药组[予恩度+顺铂(DDP)]、联合治疗组(予龙蝎消水膏联合恩度、顺铂)。各组采取相应干预措施,观察小鼠体内腹水量和腹水瘤细胞的存活率。采用RT-PCR方法,测量各用药小鼠血管内皮生长因子(VEGF)、血管内皮生长因子受体(Flt-1)、基质金属蛋白酶-2(MMP-2)mRNA的含量变化。结果中药外敷组、西药组、联合治疗组对小鼠腹水的抑制作用明显优于对照组(P<0.01);与对照组比较,中药外敷组、西药组、联合治疗组治疗后小鼠瘤细胞计数下降(P<0.05,P<0.01)。与对照组比较,中药外敷组、西药组、联合治疗组肿癌组织中VEGF mRNA、MMP-2mRNA、FLt-1mRNA的表达水平差异有统计学意义(P<0.05,P<0.01)。与西药组比较,联合治疗组的VEGF mRNA、MMP-2mRNA、FLt-1mRNA的表达水平下降(P<0.05)。结论龙蝎消水膏对小鼠移植性腹水瘤具有明显抑制作用,能明显降低肿瘤组织VEGF mRNA,MMP-2mRNA、FLt-1mRNA表达,并与恩度、顺铂具有协同增效作用。Objective To discuss the effects of ＂ Longxie Xiaoshui Ointment＂ on mice with Ehrlich ascites tumor （EAC） and to explore its mechanism. Methods Except for 10 nude mice in normal group, the other 40 mice was modeled by injection ofO. 2 ml EAC cell suspension（ 1 × 107/ml）. Then the models were randomized into four groups： control group （ normal saline）, Chinese medicine group （ ＂Longxie Xiaoshui Ointment＂ ）, western medicine group （ Endostar ＋ DDP） and combined treatment group （ ＂Longxie Xiaoshui Ointment＂ combined with Endostar and DDP）. The corresponding intervening measures were applied in each group. The inhibitory effects of ＂Longxie Xiaoshui Ointment＂ on ascites volume and EAC survival rate were observed. The contents of VEGF, Flt-1 and MMP-2mRNA were measured by RT-PCR. Results The inhibitory effects on ascites in Chinese medicine group, western medicine group and combined treatment group were better than that of the control group markedly （P 〈0. O1 ） ; After treatment, compared with control group, the EAC cell counting in Chinese medicine group, western medicine group and combined treatment group were reduced （P 〈 0.05, P 〈 0.01 ）. Compared with control group, the expressions of VEGF mRNA, MMP-2 mRNA and FLt-I mRNA were inhibited in Chinese medicine group, western medicine group and combined treatment group （P 〈 0.05, P 〈 0.01 ）. Compared with western medicine group, the expressions of VEGF mRNA, MMP-2mRNA and FLt- 1 mRNA were reduced in combined treatment group （ P 〈 0.05）. Conclusion ＂Longxie Xiaoshui Ointment＂ has inhibitory, effect on transplanted ascites tumor in mice. It can reduce the expressions of VEGF mRNA, MMP-2mRNA and FLt-lmRNA in tumor tissues markedly. It has synergistic effect with Endostar and DDP.
