摘　　要：通过分析KRT38基因的多态性和丰富程度,探讨其作为羊毛性状相关功能候选基因可能性。采用PCR-SSCP与测序技术,首次对418只中国美利奴羊(新疆型)KRT38基因的遗传多态性进行了分析。结果表明:在中国美利奴羊(新疆型)群体中,KRT38基因有AA、AB、BB三种基因型,其中AA基因型频率为0.46,AB基因型频率为0.24,BB基因型频率为0.30;A、B等位基因频率分别为0.58、0.42;中国美利奴(新疆型)羊KRT38基因P2非编码区的基因杂合度(He)为0.49,多态信息含量(PIC)为0.37,属于中度多态;测序分析发现两处碱基突变,即G85A,C95T;经检验中国美利奴羊(新疆型)群体在该位点处于Hardy-Weinberg平衡状态(P>0.05)。中国美利奴羊(新疆型)KRT38基因P2非编码区位点群体遗传变异处于中等水平,为进一步研究KRT38基因多态性与美利奴羊(新疆型)毛用性状间的相关性奠定了基础。The objective of this study was to identify the abundant genetic polymorphism of KRT38 gene and find whether the KRT38 gene could be functional candidate gene of wool trait by using PCR-SSCP and sequencing analysis in 418 Chinese Merino sheep of Xinjiang type. The results showed that KRT38 gene had AA, AB and BB genotypes in Chinese Merino （Xinjiang type） sheep. The genotype frequencies of AA, AB and BB were 0. 46, 0. 24 and 0. 30, respectively. Allele frequencies were 0. 58 and 0.42 for A and B, repectively. The genetic heterozygosity （He） of P2 non-coding region of KRT38 gene was 0. 49 and the polymorphic information content （PIC） was 0. 37 which be-longed to the moderate polymorphism. Two nueleotide mutations of G85A and C95T were found by sequencing. KRT38 gene was at Hardy- Weinberg equilibrium （P〉0. 05 ） in the Chinese Merino （Xinjiang type） sheep. The genetic variation was at the middle level in the KRT38 gene P2 non-coding region in Chinese Merino （Xinjiang type） the sheep population, and the result could be used to identify the correlation between KRT38 gene polymorphism and wool traits of Chinese Merino （Xinjiang type） sheep in the further research.
