摘  要:目的:研究指状青霉(Penicillium digitatum)提取物的致突变性。方法:采用了细菌回复突变试验,小鼠骨髓 PCE微核试验,大鼠肺及肝原代细胞程序外DNA合成试验(UDS)和大肠杆菌 K12 infa基因突变试验及 infA基因 序列测定和分析。结果:指状青霉提取物:①可明显地诱发大肠杆菌ND-160菌株回复突变;②可使小鼠骨髓 PCE微核率极显著增高(P<0.01);③可显著诱导大鼠肝原代细胞UDS(P<0.05)及极显著诱导肺原代细胞UDS (P<0.01);④可诱发大肠杆菌K12 infA基因DNA序列中5个碱基位点突变,且其中1个位点的突变还可导致翻 译相应氨基酸的变异。结论:指状青霉对原核细胞和真核细胞,无论在体内或体外均有致突变活性。Objective: We studied the mutagenicity of the extract for Penicillium digitatum. Methods: Reverse mutation assay, micronucleus test, unscheduled DNA syntnesis (UDS) in the primary cells of the lung and liver of rat and E. coli K12 infA gene mutation assay(the sequences of mutated infA gene were detemined and then analyzed by computer). Results: ①The extract induced the reverse mutation of E. coli ND-160 strain;②The extract significantly did the increase of micronucleus rate of mouse bone marrow PCE(P < 0.01);③the extract could induce UDS in the primary cells of the lung and liver of rat,and UDS was particularly obvious in the primary cells of lung (P < 0.01 );④Five points mutation in nucleotide of infA gene were induced by the extract and one of them led to amino acid variation(Lys→Val). Conclusion: Penicillium digitatum could cause gene mutation.
