摘 要:从铜冶炼厂和硫酸厂污泥中分离出常温、耐酸、异养型的硫酸盐还原菌,其还原硫酸盐的最佳温度在35~40 ℃。不同摇床转速对该菌还原硫酸盐影响不大;在20~45 ℃时培养均能在厌氧瓶中产生黑色沉淀,在40 ℃时对硫酸盐的还原量最大;在pH=5~9范围都能生长,其中pH=7时硫酸盐的还原量最大;在4 000~10 000 mg/L硫酸盐浓度下均能生长,低浓度条件下硫酸盐的还原率高于高浓度条件下;一定范围内接种量越大,硫酸盐的还原量越大;前期厌氧瓶内产气量较大,但硫化氢气体量较少,后期缓慢还原硫酸盐并有硫化氢气体产生。16S rDNA测序分析表明:该菌属于细菌界(Bacteria),后壁菌门(Firmicutes),梭菌纲(Clostridia),梭菌目(Clostridiales),消化球菌科(Peptococcaceae),脱硫肠状菌属(Desulfotomaculum),与该属中的Desulfotomaculum sp RH043和Desulfotomaculum hydrothermale strain序列同源性分别为100%和98%。该菌属于硫酸盐还原菌的一类,对酸性矿山废水治理具有一定的作用。A mesophilic,acidityphilic,heterotrophic sulfate-reducing bacteria,was isolated from the sludge near the factories of copper smelting and sulfuric acid manufacturing. The optimum temperature for sulfate reduction was 35440℃. The effect of rotation speeds on the reduction of sulfate was negligible. The bacterium cultivated at 20~45℃ was able to produce a black precipitate in the anaerobic bottle, and the largest sulfate reducing quantity can be obtained at 40℃. When the pH was 5-9 or the sulfate concentration was 4 000 - 10 000 mg/L, the bacterium can grow. The sulfate reducing quantity was the largest at pH 7 and the sulfate reducing quantity under the condition of low sulfate concentration was higher than that at high sulfate concentration. The sulfate reducing quantity can be increased with the inoculum size in a certain range. The gas production in the anaerobic bottle was large at the earlier stage, but the gas content of hydrogen sulfide was less,which was produced through the sulfate reduction at later stage. 16S rDNA sequence analysis indicated that the bacterium belongs to the Bacteria kingdom, Firmicutes phylum,Clostridia class,ClostridiaIes order, Peptococcaceae family, and Desulfotomaculum genus, whose similarity with Desulfotomaculum, Desulfotomaculum sp RH04- 3 and Desulfotomaculum hydrothermale strain in the same genus is 100% and 98% , respectively. This bacterium is a kind of sulfate reducing bacteria,and would demonstrate a good value in the acid mine drainage treatment.
